Roxarsone and Arsanilic acid are widely applied as organic arsenic feed additives in animal husbandry industry. We present a method based on hydride generation-atomic absorption spectrometry for the determination of total arsenic residues in chicken tissues, vegetables and soils.1Determination of residues of the total arsenic in chicken tissues, vegetables and soils by hydride generation-atomic absorption spectrometryWe present a method based on hydride generation-atomic absorption spectrometry for the determination of total arsenic residues in chicken tissues, vegetables and soils. A sample pre-treatment using digestion method transformed various form of arsenic into inorganic arsenic and total inorganic arsenic was reduced to the three valence state using arsenic-reducing agent, the three valence state arsenic was determined by HG-AAS. The calibration curves were linear (R2>0.99) in the range of0-10ng/mL. The coefficient of variation of with-in day(2ng/mL、6ng/mL、10ng/mL) in chicken tissues, vegetables and soils was in range of1.22%~13.21%; The coefficient of variation of day to day was in the range of1.22%-13.58%. The relative recovery was in the range of96.30%~107.95%.Limit of detection (LOD) and limit of quantitation (LOQ) were0.11ng/mL and0.35ng/mL. The method was simple and accurate enough to determine residues of arsenic in chicken tissues, vegetables and soils.2The study of arsenic residue on chicken tissues fed with arsanilic acid and roxarsoneThe purpose of this research was to estimate total arsenic content in different tissues (leg muscle, breast muscle, heart, gizzard and liver) and study the total arsenic residue regulation. The broilers were randomly divided into three groups:control group, arsanilic acid group, roxarsone group. The broiler chickens were randomly divided into three groups:control group, arsanilic acid group, roxarsone group. They were fed in three stages (1~3weeks,4~6weeks,7~8weeks). During the period of the third week and4-6week, feeds were added with recommended dose of arsanilic acid or roxarsone. During the other time, chickens were fed with basic feeds only. Chicken tissues were collected at the end of3,6,7,8week, respectively, The arsenic residues were determined by the method established in Chapter Ⅱ. After a period of continuous feeding organic arsenic, the arsenic residues in different tissues were enhanced significantly different. After the withdrawal of a week, arsenic residues reduced remarkably, after two weeks, they had the same levels with the control groups. Human healthy analysis of intake arsenic in chicken tissues:after the withdrawal time of one week, calculated by eating250g chicken tissues, human can intake inorganic arsenic at the range0.0065~0.0267mg/g, below the ADI value of0.126mg/d formulated by FAO/WTO. The experiment indicates that arsenic does accumulate in the broiler chicken tissues; the residue of arsenic in internal organs was relatively higher than in muscule. After the withdrawal time of two week, the total arsenic content met the residue relative standard.3Research on total arsenic residual of chicken manure contained with roxarsone and arsanilic acid to vegetables and soilResearch on total arsenic residual influence in the chicken manure contained roxarsone and arsanilic acid to vegetables and soil. Recommendation dose of roxarsone and arsanilic acid was added to broiler feed and collected chicken manure as organic manure in the soil for growing vegetable. A pot experiment to simulate different levels of polluted soil, total of four doses per kg soil were added chicken manure0g,5g,10g,20g respectively. And total arsenic in the samples was determined by hydride generation-atomic absorption spectrophotometry. Adding the chicken manure to the soil can enhance the total arsenic content in the two kinds of vegetables and soil, compared with0chicken manure added, other three groups total arsenic content were significantly (p<0.05) or extremely significant difference (p<0.01).Compared with spinach, lettuce easily accumulated arsenic. The soil after planting spinach had a higher level of arsenic content. Total arsenic content in edible parts and soil did not exceed the relevant standard in China. But arsenic content of the group which added the maximum amount chicken manure in spinach not edible part exceeded the standard of food hygiene.
