In this study, reproduction organs of bitches from different reproductive stage (follicular stage, corpus luteum stage and anestrus stage) was collected in slaughterhouse, the appearance and tissue structure of the uterus, ovary and fallopian tube was observed, the effects of the different reproductive stage (follicular stage, corpus luteum stage and anestrus stage) and culture mode and culture time on IVM of canine oocytes were studied. The results were as follows:1. The volume of bitches ovary from follicular stage, corpus luteum stage and anestrus stage were812.63±335.05mm3,1081.80±365.52mm3and446.03±153.89mm3, follicular stage significant higher than anestrus stage(P<0.05) and lower than corpus luteum stage(P<0.05); The weight of bitches ovary from follicular stage, corpus luteum stage and anestrus stage were0.89±0.31g,1.14±0.22g and0.71±0.18g, there were not significant difference. Fallopian tube of bitches was slender tubular organ, the length of follicular stage, corpus luteum stage and anestrus stage was11.10±1.15cm,9.23±2.12cm and5.37±1.53cm. The number of fold in follicular stage fallopian tube was more than corpus luteum stage(P>0.05)and anestrus stage(P<0.05).2. After culture for48h, the percentage of oocytes developmented to MI-MII at various stages were19.2±15.3%,26.8±21.1%,9.8±10.4%, there was not any significant difference between follicular stage and corpus luteum stage(P>0.05), but both were significantly higher than anestrus stage (P<0.05). After culture for72h, the percentage of oocytes developmented to MI-Mll at various stages were25.2±13.5%,6.6±6.7%and9.2±5.5%, follicular stage significantly higher than other two groups(P<0.05).3.After culture for48h in vitro maturation of single canine oocyte, the percentage of follicular stage oocytes developmented to MI-MII at various stages were15.9±8.0%, lower than control group(P>0.05), after culture for72h, the percentage of follicular stage oocytes developmented to MI-MII at various stages were12.4±11.3%,lower than control group(P<0.05).4.The oocyte diameter of GV, GVBD, M I and Mll were155.42μm,171.12μm,183.50μm and155.96μm, the zona pellucida wth were29.20μm,33.70μm,33.91μm and30.31μm, there was not any significant difference(P>0.05).The results of this study would to establish and perfect the bitchs in vitro maturation of oocytes training system.
