Inner Mongolia as the country’s most important livestock production base, the quality of silage getting higher and higher, while the lactic acid bacteria in silage performance directly affects the quality of the feed. To the field of silage preparation, There are great significance to filter out the fast growth rate and producing acid ability of the bacteria. The material of the test come from Tu Zuo qi High Springs Camp Ranch, Jia’s Camp Ranch and Lan Can pastures in Inner Mongolia, Green fodder samples were isolated and identified lactic acid bacteria, then the produced acid and fast-growing strains can filter out by corona electric field mutagenesis method.First step, separation of lactic acid bacteria in silage from three different sources, through the analysis of characteristics on cell morphology, colony characteristics, medium, dissolved calcium ring, etc., the medium was preliminarily identified and selected30test bacteria isolated from three different sources of silage (10per sample feed).The second step, preliminary determination of the changes of the30test bacteria’s PH in the fermentation medium, PH decreased the maximum of bacterial G-1, J-7, L-9, the degree decreased of1.62,1.68and1.57in36h, On bacterial DNA extraction, amplification, sequencing, and found the three16SrRNA sequence homology of Lactobacillus buchneri97%, They are all buchneri.The third step is to accurately measure the G-1, J-7, L-9of the growth curve and acid production. After comparison, selecting the L-9mutation to induce mutation by using the field for13KV,14KV,15KV,16KV,17KV, time of lmin,2min,3min treatment dose. From the mutagenesis field a strong16KV,17KV, time1min dose purpose strains SYL-3and the SYL-5, the final bacteria solution pH value are4.18and4.09, compared with the original strain increased by7.52%and9.51%respectively.Finally, lactic acid bacteria was separated from ABLS probiotic fermented milk, measurement parameters, explored the differences through comparing with the strains of Mutation.The test results show that the corona electric field mutagenesis of lactic acid bacteria and production of acid can improve significantly affected, the mutagenic effect of great significance to the development of silage.
