Apple chlorotic leaf spot virus (ACLSV) and Prunus necrotic ring spot virus (PNRSV) areimportant viruses infected peach trees and other fruit trees which can cause serious effect on thegrowth of fruit trees and the quantity, quality of the fruits. Cherry green ring mottle virus (CGRMV) was recently reported in China, which can affect stone fruits. These three viruses, studiedin this research, were detected by RT-PCR in order to investigate their distribution in the majorpeach growing areas in China and analysis their sequence variability. The main results are asfollowing:1. Field surveys were carried out in the major peach growing areas of China to assess thesanitary status of these three viruses. A total of417peach samples from different districts weretested by RT-PCR for the presence of ACLSV, PNRSV and CGRMV. The most frequent virus wasACLSV (83/417,20%), followed by CGRMV (12/112,11%) and PNRSV (24/401,6%).2. There are two groups of ACLSV isolates existed in peach trees.3. Coat protein sequence of PNRSV demonstrated that sequences from this study mailydistributed in clusterâ… and â…¢.4. Coat protein sequence of CGRMV demonstrated that sequences from this studydistributed sadomly.5. APCLSV was the first time reported by us in QingDao peach tree (variety:Richuanbaifeng). Comprison the isolates of APCLSV with the two sequences in GenBankdemonstrate that the sequence similarity on nucleotide level is86.3%-96.4%and93.8%-96.4%onamino acid level.Two full genome sequences of ACLSV (JN634760-JN634761) and40ACLSV-CP sequences(JN848971-JN849010) as well as one APCLSV-CP (JN873309) were deposited into GenBank. |
