| Bovine herpesvirus-1(BHV-1), a member of the Herpesviridae alphaherpesvirus subfamily,which has a double-stranded chrosome DNA,is a major pathogen of cattle.Recombinants of BHV-1expressing immunogenic foreign proteins have been reported as vaccines for other infectiousdiseases. Now days many foreign protein gene has been inserted into BHV-1as a live vectorvaccine.Rabbit hemorrhagic disease (RHD) was first observed in China1984and represents an acute,fatal disease of wild and domestic rabbits.The disease causes important economic losses incommercial rabbit production..The etiological agent is the rabbit hemorrhagic disease virus(RHDV)belongs to the Genus Lago virus within the Caliciviridae. because RHDV cannot bepropagated in vitro in cell culture systems. presently,used commercial vaccines are still preparedfrom the liver of experimentally infected rabbits.Therefore, replacing the use of infectious animalmaterial for vaccine production with new recombinant vector systems would be very desirable.We successfully constructed the real time PCR method for detecting the RHDV. Constructedtransfer vector (pGEM-gI-vp60-gEsn) contained the VP60gene expresssing box by two steps.Then the transfer vector and genome of BHV-1/gE-recombinnant on the BL cell.,After that weselected the recombinant virus (rBHV-1/VP60) on the MDBK cell by white and blue blot. Therecombinnant virus was identified by PCR,westrn blot and IFA.the immunogenicity wasconfirmed in a mice model. This research provided a technical platform for fortherimmunogenicity research of recombinnant virus in rabbit. |
