Mei, Prunus mume is an economically and nutritionally ornamental plants of the Rosaceae family. Recently, its genome sequencing project is now in progress. However, transcriptome information of this species is still scarce, with a total of around4,666Expressed Sequence Tag (ESTs) and mRNA sequences currently available in GenBank. In order to gain more insights into transcriptome, we performed transcriptome sequencing of tissues, the main conclusion as follows:1. We conducted high-throughput Illumina RNA sequencing by paired-end sequencing, according to reference transcriptome and Data Analysis, we approximately generated125,865,634high quality (HQ) reads, yielding11.3(11,327,907,060) Gb of sequence data. For each of the five samples,>75%of all reads were aligned on P. mume reference genome with a tolerance of2bp mismatches. Of the total reads,80.43%matched either to a unique (76.77%) or to multiple (3.66%) genomic locations. The remaining reads were either of poor quality or multi-mapped to the genome (6.48%).2. Based on RPKM method, the research analyse differential expression gene between tissues, and Hierarchical Cluster Analysis, we gained a number of Significantly differentially expressed in flower. Besides, Gene expression analysis revealed that,14243Unigene match to Gene Ontology database,18916Unigene match to SWISS-PROT database,14140Unigene match to KEGG pathway database,24922Unigene match to UniProtKB/TrEMBL database, greatly enriched the transcriptome of p.mume.3. Novel transcripts can be found by high throughput sequencing since present databases may be incomplete. In addition to this, we study four AS, include A5SS, A3SS, SE, RI. The result show that, A3SS is the most, with55.7%in AS and54.4%in AS gene.4. SNP analysis will help to study the variation of single nucleotide caused by the change of the protein function, and as the plum flower type of various genes with phenotypes detection basis. The Research systematically test the SNP in each organization sites.The results of this study enriched the Mei flower transcriptome, lay a solid foundation for the research of Mei molecular mechanism.
