Primula forbesii is one of the wild species of Primula L., which shares high ornamental values and landscape application potential. Plants in genus Primula are world famous alpine flowers, which are well-known for their various flower colors. As the modern distribution center for Primula, China possesses three-fifths of the Primula species in the world. Breeding parents originated from China were introduced to abroad at an early stage, thus many great horticultural varieties have been cultured and applied. Since breeding and production research in China are lagging behind, most of the valuable species are still in the wild state. According to previous research, most Primula are difficult to be introduced from their habitats to low-altitude areas. P. forbesii, however, originally distributed in lower-altitude area, can be introduced successfully. It owns favorable features such as long flowering period and pleasing fragrance. In order to breed new P. forbesii varieties with our own intellectual property, this research continued for three years focusing on the studies of floral scent analysis, cross breeding and haploid breeding of P. forbesii. Results are showed as follows:1. The fragrance of’Fragrant Luolan’cultivar in different florescences and fragrance of different cultivars of P. forbesii were collected by headspace collection and analyzed by TCT-GC/MS. The main volatile compounds of aroma were monoterpenes, esters and ketone phenols compounds. There were30principal compounds detected. The main fragrance of P. forbesii were P-linalool, a-ionone, β-ionone, methyl benzoate, methyl phenylacetate and phenol.2. Different variations of P. forbesii were separated and stabilized by using intraspecific hybridization and optimization. After3years of within-line hybridization and selection, new cultivars were bred. According to different flower colors and flower types among the large cultivated groups of P. forbesii, several lines have been divided.3. Anther culture was carried out in the research to obtain Haploid P. forbesii. Through haploid breeding, this paper laid foundations for doubling chromosomes to be homozygous germplasm in the future. Results showed that the best basic medium for anther culture was MS medium with supplements of KT (1.5mg/L)+2,4-D (0.5mg/L)+NAA (1.0mg/L), which had the highest induction. It is also indicated that activated carbon has great influence on callus induction frequency of P. forbesii. In summary, this research not only studied floral scent analysis, cross breeding and haploid breeding of P. forbesii, but also offered references for the breeding of other species in genus Primula.
