The thesis mainly researches the effect of resource restoration of Sepiellamaindroni in coastal waters of Zhejiang province. There are four respects of Sepiellamaindroni which are the piscatorial biological characteristics and the resourceevolvement, the EGFP (enhanced green fluorescent protein) tagging technologywhich is applied to enhancement and releasing, effect evaluation of enhancement andreleasing and the fishery resources management strategy were researched in thispaper.Some characteristics of Sepiella maindroni in Chinese sea area, such as lifecycle,migration, spawning, coastal growth, and resources evolution process and its reasonswere reviewed. We concluded the changes that the resource being experienced rich tobe exhausted and then be restored are all in close attach with the characteristics ofSepiella maindroni and the development activity of marine resource by human.We developed the EGFP tagging technology which is applied to enhancementand releasing to acquire the whole fishing data and cut the costs of releasing, becausethe EGFP tagging technology is better than the current tagging technology in manyrespects. The marrow of this technology depends on integrating the recombinant DNAwhich includes the promoter sequences of-actin gene of Sepiella maindroni andEGFP gene into the Sepiella maindroni genome. Then the constitutive expression ofEGFP in cells of the fertilized egg of Sepiella maindroni will happen after theintegrating. The purpose of doing this is brightening the genetically modified Sepiellamaindroni which can be tagged individuals.The current stock of Sepiella maindroni in Coastal Waters of Zhejiang Provincewas assessed via the fishing data by stationary trap nets in three areas which areLiuheng-Zhongjieshan area in Zhoushan, Dachen area in Taizhou and Dongtou area inWenzhou, which is called population density estimating. And prospect of stockenhancement and arrangement of releasing work were forecasted in the next fouryears. The number of individuals of stock assessment in three areas in2010and2011are:1.30×10~7,1.71×10~7in Zhoushan;5.70×10~6,7.07×10~6in Taizhou;3.62×10~6, 4.68×10~6in Wenzhou respectively. The stock amounts are2,794.73t、3,795.82t inZhejiang province in two years respectively. The stock of Sepiella maindroni willreach more than10,000t amounts (7.99×10~7individuals) to2.77times that of2011in2015, if the releasing number which is based on the scale of2011be fulfilled innext four years.In the end of this paper, the fishery resources management and protection strategywere researched. In the early resources recovery period, several jobs will be done,which are keeping going of enhancement and releasing, to restore and protect the habitatand spawning field of Sepiella maindroni, and to ban fishing completely individuals ofSepiella maindroni on the path of migration. We recommend that banning fishingcompletely individuals in the early resources recovery period is a great measure toalleviate the huge stress of enhancement and releasing from fishing.
