With the global climate greenhouse becoming more and more serious and animal productionbecoming more and more intensive.The heat stress in the process of animal production hasattracted more and more attention.The temperature is a key factor among which can cause thesemen quality decline.And it will directly affect the economic benefits to the livestock industry.In the process of heat stress,the heat stress proteins are a kind of high-conservative proteinswhich are produced when the body is under the influence of high temperature and other adverseconditions.And HSP70is one of the important HSPs.It can be used as a standard to evaluate thebody non-specific response under the heat stress condition.So HSP70is regarded as the goal of thestudy which is to study the different anti-heat stress additives state of the gene expression and thebody temperature response relationship.Rats are used as the animal model in the experiment and we study the gene expression ofHSP70in different anti-heat stress additives in order to lay a foundation for the study of thethermal stress on the semen quality in anti-heat stress additives mice,so150male mice are dividedinto groups according to the treat of room temperature, heat stress and anti-stress additives.Eachgroup were kept in5different periods:1day,3days,5days,7days and9days.Then we analyse theparameters of their semen quality(It include the percentage of the living sperms,sperm acrosomalintegrity rate and sperm abnormality rate).After the treatment of the experimental group,weextracted the total RNA and proteins from spermary of mice,then we used RT-PCR and Westernblot to determined the RNA and protein of HSP70.The results showed that the three anti-stress additives increased the percentage of alive semenand play a part in reducing the sperm acrosomal integrity rate and sperm abnormality rate. Theeffects of the three anti-heat stress additives on reducing semen quality of heat stressed mice aresignificantly different.Among them(P<0.05),Chinese herbal medicine is the best and the secondgroup is the electrolyte group,followed by the vitamins group.As for the expression standard ofHSP70mRNA, the anti-heat heat additives group’s expression is larger than that of roomtemperature group.But the expression amount of the anti-heat stress additives differs largely. Asfor the expression standard of heat stress proteins,the best is the Chinese herbal group and thevitamin is the lowest(P<0.05),which may be related to the effects of different anti-stressadditives.By comparing the different periods,we found the best period of different additives is alsodifferent in some degree.Therefore,this experiment is mainly to explore the best fill a prescription and amount of anti-stress additives by comparing the effects of different anti-stress on reducing the semen qualityof heat-stressed mice.It will lay a theoretical foundation for the relief of heat-stress effect on thesemen quality of economic animals and play an instructive part in the production.
