Abstract/Summary:
Voltage-dependent anion channel (VDAC) is an important protein which lies in theouter mitochondrial membrane. It is coded by a small gene family, and it not onlyplays an important role in the mitochondrial material transportation and functionalregulation, but also is involved in a series of activities in cell, such as apoptosis etc.In rice Nipponbare genome,8vdac genes were prodicted through bioinformaticsanalysis. The their expression patterns were analyzed. In this paper,3vdac genes(Osvdac2、Osvdac4and Osvdac5) were further studied. The main results are asfollows:1. The Osvdac2and Osvdac4genes were cloned and expressed by theprokaryotic expression system. Then, OsVDAC2and OsVDAC4proteins werepurified by nickel column. The respective polyclonal antibodies were prepared, thetiter is about1:20,000.2. The expression patterns of VDAC in different tissues at the different growthperiod of TYA and YTB were analyze by method of Western Blot, using the probes ofantibodies of OsVDAC3and OsVDAC5, respectively. The results showed thatOsVDAC5is differentially expressed in the different tissues at the different growthperiod of TYA, but no difference of OsVDAC5expression were observed in sametissues in YTB.3. OsVDAC5were expressed successfully in yeast expression system. Thesubcellular localization by merging of Mito-Tracker Green staining and fusion proteinof OsVDAC5-RFP under fluorescence microscopy suggested that OsVDAC5belocated in the mitochondria in yeast.
