The genetic of Musa L. mitochondrial genome belongs to paternal inheritance, and the moderate rate evolution of mitochondrial genome sequence can be used to investigate the classification and genetic relationship about banana germplasm, help us recognize the paternal relationship, enlarge the genetic base, full use of rich and wild resources, provide a theoretical basis for the new species of resistive breeding. In this study,33 wild species,9 cultivars from.Different genotypes were used,42 pairs of mitochondrial primers were selected, and sequences of ccb256, nad4L/orf25, cox2/2-3 and nad4/3- 4 sequences were selected to sequence and analysis of phylogenetic development. The results and conclusions are as follows:1)21 mitochondrial DNA primers of 44 primers can be successful transferred in banana gennplasm, proved that these mtDNA primers had the feature of good currency;2)The mitochondrial DNA had the conservative feature; that is, the slow rate of evolution of mitochondrial genome sequence can be fit to the Musa interspecies classification, the result was consistent with the morphological classification; and the phylogenetic development classification of more conservative sequences after splicing had the clear relationship with morphological classification;3)The banana cultivars were clustered with M.acuminata and M.balbisiana, respectively, which confirmed that banana cultivar was evolved from the interspecific of intraspecific hybridization, furthermore, the paternal origin could be initially speculated;4)M.yunnanensis was clustered together with the other materials of Eumusa, my advice is to classify it as Eumusa. Moreover, M.yunnanensis and M.tongbiguanensis had a close genetic relationship; they were in Yunnan province from the geographical distributions.
