Arabidopsis thaliana is an important model plant, which is widely used in plant genetics, developmental biology and molecular biology. The seed is an important stage in the life cycle of higher plants, and it is an important way to save the species. The seed germination, as the beginning of an individual life, is particularly important in plant life cycle. Seed dormancy and germination are controlled by various environmental factors and chemical substances, such as light, one of the important environmental signals, which affects plant growth and development, besides it works as the main energy source for plants, it also provide the plant with position information to adjust a number of development processes, including seed germination, seedling de-etiolation, etc.. And in addition to the light, exogenous hormone GA and ABA can also affect seed germination.FAR1 and FHY3 are very important genes in light signaling pathway in Arabidopsis, and the seed germination of the far1、fhy3、far1/fhy3 mutant plants are different from that of wild type, then these two genes may regulate seed germination, and the plant hormone GA、ABA also involved in the regulation of seed germination. In this study, by using methods of molecular biology, such as yeast one-hybrid and EMSA, we reserched the interactions between FAR1、FHY3 and the genes related to GA、ABA from the DNA aspect; at the same time, we took various seed germination experiments to research the interactions between FAR1、FHY3 and GA、ABA during the seed germination in Arabidopsis from the physiological aspect; through these experiments, we took a preliminary exploration of the reasons leading to differences in seed germination. Main research results are:Through the yeast one-hybrid experiment, we have proved that:in yeast cells, FAR1 and FHY3 proteins could combine to the FBS motif of the promoter of GA, ABA related genes, including GAR, GA2OX6, ABI5, ABR1.Through the EMSA experiment, we have proved that:FAR1 and FHY3 proteins could combine to the FBS probe of the promoter of ABA related gene ABI5 in vitro.All the germination experiments with Arabidopsis seeds of far1-2, fhy3-4, far1-2/fhy3-4 mutants and NO wild-type also indicated that:FAR1, FHY3 involved in the seed germination regulation process by GA and ABA in Arabidopsis.Exploring the interactions between of FAR1, FHY3 and GA, ABA during the seed germination in Arabidopsis, it is very important to revealing that the environmental factors regulate the Arabidopsis seed germination together with the chemical substances, by it’s signaling components interacting with the chemical substances, and it has important applications in using environmental factors and chemical substances to maintain seed in dormancy status to durable species quality of resources.
