Fluorescence correlation spectroscopy is the most sensitive methods ofsingle molecule detection technique. It is widely used in the field ofbiology and chemistry. is widely used in chemistry, biology and otherdisciplines. Fluorescence correlation spectrometer is the vector to realizethe fluorescence correlation spectroscopy technique. The paper mainlydiscusses the fluorescence correlation spectrometer hardware systemdesign, and digital correlation algorithm and data transmission system.In order to discuss the design of the hardware system, digitalcorrelation algorithm and data transmission system, the paper firstlydiscusses the basic principle of fluorescence correlation spectroscopytechnology and its realization method so as to provide a theoretical basisfor the following discussion. At first, the paper discusses core theory ofdigital correlation algorithm and realization method. And the paper usesmultichannel delay structure to calculate the correlation algorithm, anddiscusses two kinds of normalization algorithm and their standarddeviations. Then this paper begins to discuss fluorescence correlationspectrometer hardware system and determines the FPGA as the mainmicroprocessor of calculation, and make ARM as the microprocessor oftransmission system, and chooses a dual port SRAM as the data cachememory between the FPGA and ARM. Then the paper begins to discussthe design of digital correlator hardware. The system uses Verilog HDLlanguage to design hardware circuit and uses the modular method to designeach module for high speed pulse counting, correlation and data transmission. As a result, the system is designed with232channel andminimum delay period of1microseconds for correlator. Then in VisualC++development environment the system uses the MFC class library todesign computer software interface. And it uses USB to communicate withthe ARM and the computer. Then the system calculates the correlationfunction and draws curve and analyzes chemical property of fluorescencesolution. Finally, the paper compares the test results of the digitalcorrelation system with theoretical analysis and validates the digitalcorrelation theory.To sum up, this topic which is based on the fluorescence correlationspectroscopy theory and digital correlation algorithm mainly discusses thesystem hardware, digital correlation algorithm and data transmissionsystem.
