A near-infrared surface-enhanced Raman spectroscopy (NIR-SERS) method combined with multivariate analysis was developed for non-invasive liver cancer detection and after surgery patients recovery detection.In the experiment, we examined the oxyHb UV-visible absorption spectrum. Liver cancer and liver cancer after surgery oxyHb UV-visible absorption spectrum peak intensity were bigger than these heathy people, but liver cancer after surgery peak intensity were small than liver cancer. It showed the information that the Heme in the blood of liver cancer was more than heathy people and liver cancer after surgery. The Heme in the blood of liver cancer after surgery was in reducing. NIR-SERS measurements were performed on30healthy volunteers,40liver cancer and30after surgery patients which based on Ag nanofilms. Analysis indicated NIR-SERS spectra of oxyHb among healthy persons, liver cancer and after surgery patients were different. The main peak intensity showed a gradually decreasing trend among them. The relative intensity ratios between heathy people and liver cancer were the biggest difference in814、1208、1335、1587cm-1.Principal components analysis (PCA) and independent variable T test were used to diagnose liver cancer, and it get high sensitivity (86.7%) and specificity (95%). At the same time, using the discriminant analysis method to predicate group membership for after surgery patients, it could get the recovery of after surgery patients. According to assignments analysis, it showed that pyrrole ring and the group vibration mode of oxyHb had undergone a significant change. The NIR-SERS detection of oxyhemoglobin for liver cancer and after surgery patients based on PCA combined with T-test is expected to develop into a new type diagnostic tool for liver cancer and recovery of after surgery patients.
