Background The dry buds of the perennial evergreen plant honeysuckle (Lonicerajaponica Thunb.)-honeysuckle flower, has been planted in most areas of China, suchas Shandong and Henan Province. The main chemicals in honeysuckle flower includeorganic acid compounds, carbohydrates, flavonoids, volatile oils, terpenoids, traceelements and so on. The honeysuckle flower has many beneficial effects, such asbacteriostatic action, anti-virus, heat-clearing and detoxicating, cholagogue andhepatic protection, anti-allergic and anti-tumor effect, mopping up free radicals,antioxidation, boosting immunity, regulating blood lipid, easing blood pressure. Inaddition, it can treat headaches, diarrhoea, respiratory tract infection, fever, etc.Meanwhile, it also has obvious therapeutic effects towards cardiovascular disease.Flos Puerariae, as the flower of medicinal and edible plant pueraria, native mainly toHenan, Hunan, Guangxi, Sichuan, Anhui located in China. The major effectivecomponents of Flos Puerariae are flavonoids, volatile oils, saponins, sterols, alkaloids,amino acids, etc. Flos Puerariae has sober up and attendant liver function and it canlower blood sugar, blood-fat, protect the cardiac muscle, anti mutagenesis, preventallergies, improve the learning and memorizing abilities and something like that. Thehoney has been regarded as a tonic and of medicine food homology since ancienttimes. Its primary chemical constituents consist of carbohydrates, mineral salts,proteins, amino acids, trace elements and so forth. The honey not only has protectiveactions of liver, cardiovascular system, gastrointestinal system, but also has promotingeffects in healing diseases of nervous system and respiratory infections. Today theresearch on anti-inebriation effects of Flos Puerariae has been matured, however, thesame effects of honeysuckle flower has not been reported, let alone the functionalbeverage with honeysuckle flower, Flos Puerariae and honey as main ingredients.Purpose To study the effects of alleviating alcoholism of honeysuckle flower and Flos Puerariae via animal experiments. To prepare functional beverage based onhoneysuckle flower and Flos Puerariae by the orthogonal experiments. To makeprocess design of functional beverage in order to provide the basis for actualproduction by means of technology testify.Method For animal experiments, firstly, male Kunming mice aged2months providedby animal experimental center of Shandong University were randomly assigned to8groups at10each weighed23~26g. Control group, high dose group of honeysuckleextract, middle dose group, low dose group, compound liquid group1, compoundliquid group2, compound liquid group3, and compound liquid group4. Each groupwas given the corresponding solution by gavage for0.25mL/10g body weight,30mins later, each group was given White wine (56o) by gavage for0.15mL/10g bodyweight. Then the latent periods of drunkenness, the time of sleeping and the time forsoberness of each group mice were recorded. Draw conclusions via data analysis. Andthe blood concentration of alcohol in optimal proportion of compound liquid chosenby control group, honeysuckle extract, Flos Puerariae extract, and righting reflexexperiments was determined.The functional beverages using honeysuckle extract, Flos Puerariae extract andauxiliary materials by orthogonal experiments were prepared, the best formula waschosen via sensory analysis and the active ingredient concentrations of honeysuckleand Flos Puerariae were assayed used in experiments.The technological process chosen in the task was made for demonstration andmaterial balance so as to confirm if the functional beverages studied deserved actualproduction.Result Compared to the control group, the rate of alcoholism of groups withhoneysuckle flower of the low, middle, high dose, compound liquid groups1,2,3aswell as4decreased in mice and of them, compound liquid group2had the mostobvious effect; the tolerance time for drunkenness of the low, middle, high dosegroups of honeysuckle flower, compound liquid groups1,2was lengthened and allthe groups came to extremely significant differences(P<0.01), compound liquid group4had marked difference(P<0.05)while compound liquid group3showed nosignificant difference(P>0.05); The time for soberness was shorted sharply amongthe low, middle, high dose and compound liquid groups1,3and4of honeysuckleflower and it decreased progressively with increase of honeysuckle flower’sconcentrations but without marked diversities(P>0.05).Compared to the control group, concentrations of ethanol in blood changed asfollows: concentrations of ethanol in the test groups with honeysuckle flower werehigher between30and60mins while it was opposite with extremely obviousdifferences after90mins in the honeysuckle flower experiments; The results were thesame as described above in the Flos Puerariae tests; Concentrations of ethanol in theoptimal proportion of compound liquid group were lower with extremely obviousdifferences in every time quantum after feeding white spirit for30mins; Compare thehoneysuckle flower experiments, the Flos Puerariae tests and the optimal proportionof compound liquid group, concentrations of ethanol in the optimal proportion ofcompound liquid group was lower than the other two groups with extremely obviousdifferences.Conclusion Honeysuckle extract could decrease the drunkenness rates of mice,extrend the latent periods of drunkenness and decrease the time for soberness,however. Honeysuckle extract and Flos Puerariae had the most marked effects on theethanol in mice when the ratio was3:7. Experiments that the honeysuckle flower andFlos Puerariae had synergies effects in decreasing the rate of mice intoxicated.The optimal formula of functional beverage based on honeysuckle and FlosPuerariae was honeysuckle extract20.17%, Flos Puerariae47.06%, honey15.73%and citric acid0.10%. The content of chlorogenic acid in honeysuckle flower used inthe experiments was3.085%; The total isoflavones in Flos Puerariae used in the testswas7.14%.The functional beverage based on honeysuckle and Flos Puerariaedeserved actual production by technology testify, material balance and economicaccounting.
