RNA interference is the widely self-protection phenomenon in eukaryoticcells.It can antagonize the exogenous gene such as viral genes、artificialloaded genes against and can also degrade the mRNA which abnormal genetranscription also.When the exogenous genes integrate into the host cellgenome randomLy and the attachment to the host cell transcription,thereoften produce some double-standed RNA. The host cell then rapidly producesreactions of these double-stranded RNA. Dicer RNase L in the cytoplasm canmake double-stranded RNA to cut the small fragments of RNA wia particularlength and structureth,namely siRNA.These siRNA is complementary to themRNA of the exogenous gene. Therefore the compound can mediate a seriesof enzymatic reactions. for example, firstly RNA induced by the silencingcompound, then combined with mRNA that expressed by exogenous genesand gradually induce a series of mRNA degradation reactions of the host cell;Finally, protein expression of the exogenous gene is disappeard. Thismechanism occurs in the post-transcriptional. It is also said that occurs themodification or translation of the mRNA level.It is lasting、specific and efficient.So this mechanism is not only well suited for analysis of unknown genes inthe genome era,but also for the clinical gene therapy opening up a new path.With RNA interference technology gradually maturing, Peple also recognize that RNA interference carrier is the key of this mechanism whethercan be widely applied. RNA interference carrier can be divided into twocategories,i.e.,viral carrier and non-viral carrier. The viral carriers areconstructed according to a certain structure itself and transfection mechanismof the virus with higher transfection efficiency. But the preparation is morecomplicated with highly immunogenic and non-oriented. Non-viral carriertransfection efficiency is low,but with the security,low immunogenicity,andlow toxicity characteristics. Therefore,to find a high transfection efficiency butalso has security features transfection carrier is a research hotspot of RNAinterference.This paper attempts to select FF dipeptide as raw materials and usesupramolecular self-assembly technology to make it to be with regular tubularand spherical nanomaterials. This peptide-based material is low toxicity to cell.After further experimental verification,the transfection efficiency is higher thanthe the common carrier lipo2000.Therefore it is a new、efficient、non-toxic RNAinterference carrier.This paper lays the foundation for the further gene therapy of RNAinterference. We believe that the phenylalanine dipeptide nanomaterials canplay a role as a novel RNA interference vector. Compared with the existingRNA interference vector,they are smaller cell toxicity、higher stability、andhigher transfection efficiency. They are both obvious advantages and broaddevelopment prospect.
