| Perilla is a kind of medicinal and edible herb plant which is annually grew all over the country. Exploiting the perilla resource possesses great significance both in utilization of perilla and its prospective application in other fields such as food, pharmacy, and cosmetic industries. With perilla as raw material, our research and results are mainly stated as follows:We initially conducted experiments to choose the optimum extraction solvent for extracting perilla anthocyanin and determine the adsorption variables for purification of anthocyanin by means of XAD-7. The result showed that the optimum extraction solvent was mixture solution comprising of ethanol-water (40%ethanol) and acid-water (6%acetic acid), and the adsorption process parameters were as follows: adsorption conditions applied concentration of anthocyanin in sample solution (0.26mg/mL), processing volume (4.7BV), flow rate (2mL/min), and temperature (25±2℃), and the adsorption conditions applied eluent volume (3BV, ethanol (100%, v/v)) and flow rate (2mL/min). Soluble sugar and protein impurities could be removed and the recovery rate was92.4%.We used HPLC-MS to analyze the main components of perilla anthocyanin and its stability finding out that malonylshisonin was the main components. Perilla anthocyanin was water-soluble, was able to withstand a short time of heating under high temperature, and its color varied with the change of pH. The dark environment and low temperature was the best storage conditions. The metal ion, food additives, oxidation and reductive substances could affect the stability of the perilla anthocyanin.We succeeded in extracting flavonoids substances from perilla waste which had undergone extraction of anthocyanin, and determined that the main flavonoids compounds were Apigenin7-O-caffeoylglucoside, Scutellarein7-O-diglucuronide, Luteolin7-O-diglucuronide, Apigenin7-O-diglucuronide, Luteolin7-O-glucuronide, Scutellarein7-O-glucuronide. Perilla flavonoids had a good resistance capacity to oil oxidation effect and the effect on the inhibition of lard oxidation was superior to rapeseed oil. Its oil oxidation effect was the best when it applied combination of perilla flavone (0.2%) and citric acid (0.05%). Besides, the perilla flavone had an effective capacity of anti-allergic, blocking nitrosamine, and clearing the nitrite. |
