This test detects the microbial pollution in sheep slaughter first. Take the GB/T4789.2-2010《Food microbiology examination-Determination of the total number of colonies》、GB/T4789.3-2010《Microbiological examination of food coliforms》 as a reference, expect to provide a basis for slaughtering enterprises in health regulators in the slaughter and processing. The results showed that the surface average total number of colonies by traditional method and standardize method of slaughtering sheep carcass were3.54Logcfu/cm2and3.36Logcfu/cm2; the result of average coliform by traditional slaughtering and normative slaughter were2.84LogMPN/cm2and2.60LogMPN/cm2; The aerobic bacterial count and coliform of sheep carcass that after cleaning in normative slaughter were reduced to3.13Logcfu/cm2and2.47LogMPN/cm, which shows surface microbial contamination degree before and after the carcass cleaning process is significant difference (P<0.05). It also shows that, even Specification slaughterhouse, the slaughter and processing remain polluted to varying degrees.This test also takes a more systematic study on quick detection of common pathogeniccbacteria, gets Staphylococcus aureus and Salmonella as the purpose of strain. Exploring one method to detect S.aureus and Salmonella by duplex PCR. Through the design of primers, primer-specific validation and optimization of primer in the amplification system, the Taq enzyme, dNTP, annealing temperature and other factors to establish a rapidtesting program, The result is10×buffer2.5μL, MgCL22μL, template is2μL, dNTP2μL, the primer of Staphylococcus aureus is0.7μL、the primer of Salmonella is0.5μL, Taq enzyme is0.6μL, the rest up by deionized water to25μL; the augmentation program is94℃of initial denaturation for5min,94℃of degeneration for30s,60.2℃of annealing for45s,72℃of extension for1min, get35cycle, and72℃of last extension for5min。Then detect sheep carcass surface microbial samples and mutton samples of microbial, take 《GB/T4789.10-2010food microbiology test-Staphylococcus aureus》、《GB/T4789.4-2010microbiological examination of food-salmonella test》 as reference. The results showed that the duplex PCR detection method with high accuracy.
