The Propolis resin-like secretions of bees from plant buds hold and branches collected and mixed with the complex substances formed by the bees on the pieces of gland secretions and beeswax, its main chemical composition of the flavonoids, mushroom vinyl, phenolic acids compounds and other biologically active substances, have antiviral, antibacterial anti-inflammatory, anti-tumor, antioxidant, lowering blood pressure, regulate immune function and so on. In recent years, Propolis is widespread concern on a global scale because of its variety of health care and adjuvant therapy.This paper focus on Propolis, the optimum extraction technology of flavonoids in which purified total flavonoids of propolis extract and purified propolis extracts for antioxidant and antibacterial efficacy study.The test conclusions are as follows:(1)An ethanol extraction, by a single factor analysis and according to the center combinations (Box-Behnken) experimental design principles, the results show that the extraction time10min,82%of the ethanol concentration, extraction temperature of72℃. Under these above conditions, Propolis total Flavonoids rate was22.88%.(2)Ultrasonic extraction, through the analysis of single factor and center combination (Box-Behnken) experimental design principles, the results show that27min of ultrasonic time, ultrasonic power220w, the ethanol concentration of73%, extraction temperature66℃. Under these above conditions, Propolis total Flavonoids rate was23.38%.(3)Microwave extraction method, through the analysis of single factor on the basis of the analysis of single-factor test, according to the center combinations (Box-Behnken) experimental design principles, the results show that the microwave time of70s, microwave power of282w, the ethanol concentration80%, extraction temperature was77℃. Under these above conditions, propolis total flavonoids was25.41%.(4)Purified Propolis for flavonoids purification the experimental selected macroreticular resin as adsorbing material to determinate different concentration gradient elution liquid propolis flavone content by column chromatography. The results shows, firstly to use the water to wash the soluble impurities, using30%ethanol water system as the mobile phase can separated propolis flavone and other components, so as to get the purpose of purification from the other compositions.(5)Total flavonoids of propolis extract in vitro antioxidant capacity determination, the results show that: as flavonoids concentration is0.25mg/mL, the clearance rate has reached98.3%; as flavonoids concentration is0.05mg/mL,the clearance rate of DPPH has reached96.9%; the clearance rate of the hydroxyl has reached98.9%; the clearance rate of the NO2-ion has reached97.9%. (6)Purified propolis extracts antibacterial experiment, The results shows, four kinds of food caused by rot fungus for antibacterial experiments.
