Securinine is the major alkaloids of Euphorbiaceaeplants, suffruticosa(Pall.)Rehd Securinine is kind of GABA specific receptor antagonists, which exerts the significant role of making the central nervous system exciting and has become an important clinical neurological medication and commonly used medication. The latest research results show that Securinine can markedly improve memory, which has a huge potential development of drugs for preventing and treating (Alzheimer Disease, AD). In addition, domestic and foreign scholars find that Securinine also has wide range of biological activities for the resistance to malaria, antibacterial, antiviral and the like. Therefore, in recent years, researches about Securinine are growing fastAlthough people have attached great importance to the effect of Securinine, its extraction technology is still backward. Recently organic solvent for extraction is often applied. The approach of extraction is simple, but it has great disadvantages, for instance, huge organic solvent consumption, trival extraction process and long cycle, serious environmental pollution, low product yield and purity, etc. It seriously hindered the industrial production of Securinine and the further research and development of other functions. This paper adopts the approach of linking the relevant formulation nodes of traditional acid water soaking extraction formulation, ion exchange formulation and Saxhlet smoking formulation, which effectively optimized the extraction of Securinine, further improve the yield and purity of Securinine and validates whether Securinine has the organic activity of anti-fatigue through animal experiment. The specific research contents and results are as follows:First, the technology of linking traditional acid water soaking extraction formulation and ion exchange formulation is applied to extract Securinine from branches and leaves of Securinine plants. After a single factor experiment and Box-Behnken Design method and using Design-Expert software to conduct quadratic regression analysis to experiment results, the optimal parameter of production technology is finally determined:the rate of liquid material is19:1, water bath temperature81℃, extraction times the three times, ethanol concentration89%, extraction rate is0.48%and the purity of Securinine91.5%. Thus it effectively optimizes the extraction process of Securinine.Second, mice are used to establish animal anti-fatigue model. The animal experiment results show that Securinine has no significant effect to the weight of mice; low dose and moderate dose of Securinine can obviously extend the time of loaded-swimming and time of turning sticks, can at the same time significantly reduce blood urea nitrogen content of the mice after its moving and can effectively improve the content of hepatic glycogen. It has the effects of enhancing the moving endurance of mice, increasing the substance reserves of glycogen and speeding up the urea nitrogen of body’s metabolism. Results show that the anti-fatigue activity of Securinine is very obvious, and this effect is enhanced as intraperitoneal injection doses are improved.
