As a kind of medicinal and edible fungus, ganoderma lucidum has many functions inmedical care. In ganoderma lucidum, ganoderma lucidum polysaccharide is one of the mainactive components and play a important role in health care. Lactobacillus bifidobacteriumis an important human intestinal flora which is closely related to people’s health. However,accompanying with the increasing of growth and the changing environment, the amounts ofL. bifidobacterium spp. become less and less. Bifidus factor is a kind of material which canpromote the proliferation of bifidobacterium. Through the vitro replenishment of bifidusfactor products, it can promote the proliferation of bifidobacterium in intestine.The ganoderma lucidum is treated as the raw material to research the extractiontechnology of ganoderma lucidum polysaccharide. With ascorbic acid (hereinafterabbreviated as Vc) as the contrast, the antioxidant activity of ganoderma lucidumpolysaccharide was studied. At the same time, proliferation effect on the bifidobacterium ofganoderma lucidum polysaccharide had been studied. Then, the ganoderma lucidumpolysaccharide is applied to milk production to develop a health-care beverage. Through theexperimental study, the conclusions are as follows:(1) Cellulose enzymatic extraction, cooperated with microwave pretreatment andultrasonic synergy was used for extraction of ganoderma lucidum polysaccharide. Based onthe single factor experiments, designed a trial of four factors and three levels. It had shownthat the test program by the response surface optimization analysis combined with the actualtest condition. According to Box-Behnken design principle, the optical parameters ofextraction of ganoderma lucidum polysaccharide was obtained as bellow: microwave poweris539w, microwave pretreatment time is120s, pH value is5.0, solid-liquid ratio is1:30,ultrasonic power is225w, cellulose ratio is2.2%, leaching temperature is55, leaching timeis52min. Under such situations, polysaccharide yield can be up to16.43mg/g.(2)Ganoderma lucidum polysaccharide has a certain reduction ability of free radicals. In a polysaccharide concentration ranging from0.2to0.4mg/g, reducing power increasedwith the increasing of polysaccharide concentrations. Ganoderma lucidum polysaccharidealso has a certain ability to remove DPPH and·OH. The IC50(half inhibition rate) ofremoving DPPH was as many100times as Vc, and the IC50of removing·OH was about60times, while the IC50of removing O2-· was as the same as Vc IC50, which showed thatganoderma lucidum polysaccharide has a strong ability of removing O2-·.(3)When ganoderma lucidium polysaccharide was added to youth bifidobacteriumbasic culture medium as the amount of0.08%, the growth youth bifidobacterium had noobvious lag phase, speeding up the growth rate. The dilution method experiment results hadshown that: youth bifidobacterium viable amount was1.38×109after18h in anaerobiccultivation environment, up to5.28times than in the normal control group. Saving at4℃after two weeks, the survival rate of youth bifidobacterium was30.8%, meaning1.7timesof that in the normal control group. Both experiment index are similar to the results ofpositive control trial group which was added fructo-oligosaccharide. Therefore, ganodermalucidum polysaccharides can be used as a bifidus factor to applying in the cultivation ofbifidobacterium.(4) The ganoderma lucidum polysaccharide was added as a functional factor to themilk beverage productions. Through orthogonal test, the best ingredients formula wasobtained as follows:13%white granulated sugar,4%dried skim milk,0.03%potassiumsorbate,0.2%citric acid,0.2%lactic acid,0.06%citric acid sodium,0.08%ganodermalucidum polysaccharide. The product has a beautiful flavor, and delicate texture, showingsweet and delicious taste.
