Immune active peptide is an extre me ly important bio logical active substance,which had strong stability, high bio logica l activity, can strengthe n the bodyimmunity,a nd improve the ability to fight infection,reduce the inc idence of adisease, and other important biolo gical activity. The topics selected alka lineprotease to hydrolyze defatted wheat germ protein, it was found that thehydrolysates with good immune activity by anima l immune test.The ma in researchcontents and results were as follows:(1)The best cond itions of the defatted wheat germ protein extraction processwere researched, a certain of α-amylase was added in the traditio nal method ofdissolution with a lkali and deposition with acid and it can evidently increase theyield ratio of wheat germ protein.The wheat germ protein extraction technologywas optimized with the Box-Behnken centra l compos ite design and responsesurface methodology simultaneous ly;The results ind icated that the best extractioncond itions were: Contro lling the amo unt o f water (15times of that of rawmaterial),pH9.5of a lkali d issolution,which would go on47min.(2)The alka line protease was chosen to hydrolyze wheat germ protein in thetest. The optima l conditio ns was obtained by s ingle factor and orthogona lexperiments as follows:pH8.5,temperature50℃,enzyme quantity0.4Au/g,substrate concentration1%and hydrolysis time150min,under whic h the degree ofHydrolys is of the reactio n reach35.62%.(3)Using ultra filtration techno logy for preliminary separatio n of wheat germhydrolyzate,and the adsorption and purification of the hydrolysates was operatedwith DA201-C macroporous resin.The results show that the DA201-C macroporousresin has a good adsorption performance to hydrolysates,the optimum injectionrate and concentration was1BV/h, and50mg/mL. The most appropriateconcentration of ethano l elutio n and elution rate was75%and2BV/h,the highestwashout rate of the hydro lysates was up to96.39%.The hydro lysates desalinationrate and recovery rate can reach85.90%,73.25%after purification.(4)The immunity adjustment effect of hydrolysate that alka li protease to thewheat germ protein were stud ied.Classified to the wheat germ polypeptide us ingultrafiltratio n me mbrane of3000Da, mice received with polypeptide by gavage fortwenty-eight days.After that, immune organ,the content of mura midase andimmuno globulin in mice were determined and their immunologica l activities wereevaluated.The results showed that wheat germ polypeptide of mo lecular we ight﹤ 3000Da was the strongest on the immunolo gical activity,the second was molecularweight﹥3000Da;Wheat germ prote in had the weakest effect on immunity....
