Optimization Of Fermentation Process For Pleuromutilin

Pleuromutilin is the synthetic precursor of many antibiotics, studing on its many derivatives and applied in practice in sixty years when Pleuromutilin was found. Antibiotics belonging to Pleuromutilin have special characteristic of no pollution and no residual to the environment and anti-microbial mechanism. The derivates also have difficulty in producing drug resistance and apply in broad prospect. In order to improve potence、availability of equipment fermented level and reduce costs, this study contains the optimization of seed medium、condition of fermentation、fermentation medium and way of fed-batch.First, every constituent of seed medium is studied through single factor test to observe the influence of seed vitality. Second, selected four medium components which have more effect are tested through orthogonal experimental design. Last, Potence of Pleuromutilin as index, medium components are certained that is consist of17g/L glucose23g/L soybean cake power、1.5g/L KH2PO4、0.075g/L Ca(NO3)2、0.5g/L MgSO4、0.15g/L NaCl、0.05g/L FeSO4.In this condition, strain have characteristics of high vitality,growth indicator and potence.In addition, every constituent of fermentation medium is stμdied through single factor test.Then selected three medium components are tested throμgh the Design-expert8.05to response sμrface analysis. At Last, Potence of Pleuromutilin as index, medium components are optimized for40.1g/Lglucose、33.76g/L corn steep liquor、5.307g/L bean oil、6.0g/L soybean cake power、1.5g/L CaCO3、0.40g/L MgSO4、0.04g/L FeSO4. With improvement of the condition, fermentation index raise significantly.Finally, according to institution of fed-batch, there are two different methods about fed-batch, which have little difference in fermentation total. But second method is better than first method, because of consumpting less fed-batch. As a result, save the fermentation costs and enhance the level of fermentation.
