Resveratrol is a natural anti-oxidants, it can reduce blood viscosity, inhibit plateletcoagulatie and vasodilation, maintain blood expedite and possess other functions, the scope ofapplication of resveratrol increasingly widespread. This paper set up the UVspectrophotometry, bromometry and the molecular fluorescent methods to detect resveratrolcontent and studied the extraction resveratrol from grape waste residue and the besttechnological conditions of seperation and purification resveratrol using macroporousadsorption resin.When detecting the resveratrol content by the UV spectrophotometry, the solution shouldbe acidic medium and the detecting wavelength λ was306nm, the relationship betweenabsorbency and concentration was A306=0.1169c(μg/mL)+0.0064; when using the bromometry,it concluded that the dosage of2mol/L HCl and0.1mol/L bromine reagent separetly were1.00mL and5.00mL, and under that condition, one molecule resveratrol could react with4molecules bromine reagent; and when by the molecular fluorescent method, the best detectionconditions were adding no buffer solution in the solution, the dosage of5%AlCl3was1.00mL,the static placing time was20min and the motivate and emission wavelength were312nm and406nm.The optimizing technological conditions of extracting resveratrol from grape wasteresidue were studied in this paper.We researched the pH, the ratio of solid to liquid, thetemperature and the extracting time having the effect on extraction volume by the singlefactor experiments, and on that basis, through designing the orthogonal experimental, andconfirmed the best extraction conditions: the ratio of solid to liquid was1:35, pH was4, thetemperature was40℃and the extracting time was6.0h, and the once extraction volume was1.204mg/g; Besides, detected the resveratrol content was69.53μg/mL by the bromometrymethod.The adsorption and desorption performances the LS-303macroporous adsorption resin toresveratrol using the static and dynamic adsorption and desorption experiments wereresearched in this paper, as well as the adsorption quantity, adsorption dynamics and theabsorption thermodynamics performances to resveratrol caused by the LS-303macroporousadsorption resin, and the influence of pH, resveratrol concentration and other factors on theadsorption rate. The experimental results showed that when the pH was2to8, the LS-303macroporous adsorption resin showed a good adsorptivity to resveratrol. Under the experimental conditions, the balance time of the static adsorption was60minutes. Theisothermal adsorption fit for the Freundlich and Langmuir equations and the adsorptionprocess were spontaneous and thermodynamics exothermic. With the length of the adsorptioncolumn was20.0cm, the diameter was1.5cm and contained15.00g LS-303resin in it, theabsorption rate of resveratrol solution (50mL) reached up to99.45%, whose flow rate andconcentration were5.0mL/min and20μg/mL, the resveratrol which was adsorbed could becompletely cleared with80%ethanol solution.The seperation and purification resveratrol extracted from grape waste residue also wasstudied in this paper.After separating and purifing the resveratrol extracting solution by theLS-303macroporous adsorption resin, the resveratrol concention was25.45μg/mL, and thepurity increased obviously, and the yield of seperation and purification process was72.9%.After concentrated the resveratrol solution which was separated and purified by the adsorptionresin got45.81μg/mL resveratrol water solution; the total yield of extraction, seperation andpurification and concentration processes was65.6%.
