Abstract/Summary:
Herbal Blitzkrieg Extractor(HBE) based on Smashing Tissue Extraction(STE)has the superiorities of being operated under normal temperature, fast and effective,wide application and so on. Liquid-solid circulating fluidized bed (LSCFB) has beendrawing attention for its continuous operation, fast mass transfer, well contactbetween liquid phase and solid phase, ability to deal with the feed with particles. Theextraction and isolation of flavonoids in Ginkgo biloba leaf by combining HBE withLSCFB was proposed and investigated in the paper, including the way of removingimpurity and the optimal condition of HBE, followed by the adsorption isolation ofHBE extract with AB-8macroporous resin in LSCFB. In addition, the technologicalcondition of isolating GA was studied, which was necessary to the appliciton of thecombined method in extraction and isolation of Glycyrrhizic acid (GA).Results indicate that lipid soluble impurities can be removed by evaporating theextract to the1/3of the original feed in volume under reduced pressure. The optimalextraction process is: solid-liquid ratio (g/mL)1:20,70%ethonal,120V,4min. TheHBE extract was purificated by the AB-8macroporous resin LSCFB(semicontinuousoperation), and the results reveal that the adsorption and the desorption rate wererespectively63.12%and84.44%. So it is feasible to extract and isolate the effectivecomponents in Chinese traditional medicine with the method of HBE combined withLSCFB.The method of determining the content of glycyrrhizic acid in licorice extractumwith polysaccharide extracted was established in the paper. The optimal reactivecondition: color development reagents are2.5mL6mg·mL-1vanillin and2.0mL75%(φ) H2SO4, temperatureis75℃, reactive time is25min. The method presentsgood precision (RSD=4.39%, n=6), stability (RSD=1.09%, n=60) within1h and thehigh average recovery (X=95.93%, RSD=4.62%, n=5). So it is reliable to determinethe glycyrrhizic acid in licorice extractum with absence of polysaccharide components.The non-polor macroporous resin X-5is favourable for the adsorption of Glycyrrhizicacid according to the screen test. The results reveal that adsorption effect is better inthe condition (original pH,1.75mg·mL-1sample concentration,35℃, oscillation at constant temperature and slow rotate speed), and60%ethonal (0.5mL·min-1desorption flow rate) is proper to the desorption process.
