| Aminoglutethimide3-(4-aminophenyl)-3-ethylpiperidine-2,6-dione had beenapproved under the trademark Remenon for the treatment of anticancer drugs. As amedicine for anticancer, it was developed by Novartis in Switzerland in1955.Aminoglutethimide is still under the age of patents by now. So it is necessary toexploit an economical and practical route.This thesis includes three chapters:Chapter1: The development in syntheses of AminoglutethimideIn this section, different synthetic methods of aminoglutethimide are reviewedbased on the literature.Chapter2: The syntheses of racemic AminoglutethimideIn this section, key intermediates,2-ethyl-2-phenylpentanedinitrile and methyl4-cyano-4-phenylhexanoate derivatives, for synthsis of aminoglutethimide have beenprepared under phase transfer catalysis for the first time. Furthermore, racemicaminoglutethimide was synthesized according to the literature procedure using theabove intermediates.Chapter3: The syntheses of chiral AminoglutethimideThrough Michael addition reaction, the main chiral intermediates of aminoglute-thimide was got by using the chiral phase transfer catalysts. For the first time, Evansauxiliary agents was used in Michael addition reaction to control stereoselectivity toget chiral aminoglutethimide. |
