Huperzia Serrata is a kind of traditional Chinese herbal medicine, Chinesescientists extract the Huperzine A from it in the1980s. Huperzine A is a kind ofalkaloid as an effective natural AChE inhibitors, which used to treat senile dementia,myasthenia gravis, memory recession and other diseases.Firstly, the chromatographic conditions of determinate Huperzine A content byHPLC were established. Then preliminary experiments of extraction Huperzine A werecarried out. The best condition as follows: use1.5%tartaric acid as solvent, extractingtime was24h, two extracting times. In the paper four parts experiments were processedas follows:(1)The technology of chitosan clarification Huperzia Serrata extracting was studied.Firstly, the influences of different single factor on clarification effects were investigated.The extraction technology was optimized by orthogonal test based on single factorexperiment. The optimal conditions were as follows: the ratio of solid-liquid of1:10(g/mL), the pH value of5, the reaction temperature of50℃, the concentration ofthe chitosan of0.5g/L, the stirring time of60min and the stirring rate of100r/min.According to the conditions mentioned above the luminousness can achieve81.6%. Theverification experiment results indicate that added clarifying agent will not affect thetotal weight of Huperzine A. It also can improve product purity and reduce the heavymetal content.(2)The adsorption capacities and the desorptions of five different resins towardsHuperzine A were studied. The results showed that C004cation exchange resin exhibitsthe best effects. The optimal technological parameters of C004cation exchange resinwere as follows: the pH value of sample solution3.0, flow rate3BV/h,3mol/Lammonia water-80%ethanol of elution, elution volume3BV, elution velocity1BV/h.Validation experiment results show that the reproducibility of this method is good.Under the optimum conditions, the content of Huperzine A was up to5%.(3)The technological conditions for extraction Huperzine A by supercritical fluidextraction were studied. The optimal conditions were as follows: with the raw materialalkaline pretreatment by ammonia, use200mL95%ethanol for entrainer agents,extraction temperature35℃, extraction pressure20MPa, extraction time180min, underthe optimum conditions the extraction rate of Huperzine A was up to11.44%. Finallyuse supercritical fluid extraction from semifinished product which Huperzine A contentwas0.634%was studied. The results show that the highest extraction rate was16.74%and the purity of Huperzine A in the product can reach18.15%.(4)The pilot scale experimentation of5groups about Huperzine A extraction wasprocessed based on the data of lab trials. The results show that the average extraction was83.4%and the average purity was4%. Then summarized the problem whichappearing during the pilot scale experimentation and proposed the solutions, anddiscussed the“three wastes”treatment problems. Finally has also carried on the simpleanalysis to its technical economy, the total cost of every production cycle is¥27300,and the net profit is¥7200. The results show that the process both in technological andeconomical are feasible, and can be used in industrial production.
