Now many pharmaceutical wastewater was treated with biological treatment process, thestudy of enzyme activity which catalyzed organic matter degradation and the diversity ofmicrobial community structure of activated sludge process was significant to improve theeffect of activated sludge process. This paper studied the influence of the different conditionsof influent7-ACA concentration, DO concentration, pH, temperature on treatmenting7-ACAwastewater with NMBR and static test device, analyzed the enzymatic activity of importantenzymes and the drug-fast by producing β-lactamase and the diversity of microbialcommunity structure of activated sludge microorganisms at the stable period under everycondition.The removal efficiency of organic pollutant at different concentration of influent7-ACAand DO, different pH and temperature: the removal rate of7-ACA was the best when theconcentration of influent7-ACA was30mg/L; when the DO concentration was2.5~3.5mg/L,the removal rate of COD and BOD and7-ACA were the best, COD removal rate was94.4%,BOD removal rate was97.3%,7-ACA removal rate was47.6%; The optimal pH andtemperature treating7-ACA wastewater were8and35℃respectively.The highest value of enzymatic activity corresponding to different influent concentrationof7-ACA: urease, SOD-0mg/L; dehydrogenase, nitrate reductase-50mg/L; invertase-40mg/L;PPO-100mg/L. The highest value of enzymatic activity corresponding to the different DOconcentration: urease-0.5~1.5mg/L; dehydrogenase-2.5~3.5mg/L; invertase-3.5~4.5mg/L;nitrate reductase, SOD, PPO-4.5~6mg/L. The highest value of enzymatic activitycorresponding to the different pH: urease, dehydrogenase-11; invertase-4; nitrate reductase-3;SOD-10; PPO-7. The highest value of enzymatic activity corresponding to the differenttemperature: urease-23℃; dehydrogenase, SOD-40℃; invertase-50℃; nitrate reductase,PPO-45℃.With the increasing of the concentration of influent7-ACA, microorganism producedβ-lactamase which stimulated by the7-ACA, the activity of β-lactamase increased first andthen decreased. When the7-ACA concentration was30mg/L, β-lactamase activity was thehighest1212.9U, the removal rate of7-ACA was the best. When the microorganism synthetized β-lactamase, the optimum pH, temperature, DO concentration range was8,35℃,2.5~3.5mg/L respectively.Finally this paper analysed the diversity of microbial community structure at thedifferent environmental conditions. With the increase of the concentration of influent7-ACA,Uncultured bacterium clone LR A2-3and Propionibacteriaceae bacterium WN033etc.gradually disappeared, Uncultured bacterium clone A199and Uncultured Alphaproteobacteriabacterium etc. gradually produced, these micro-organisms affecting by the changingenvironmental conditions generated adaptive mechanism due to emerge drug-fast to the7-ACA wastewater, finally they becomed the important component of the microbialcommunity at the stability stage; With the increase of temperature, Dechloromonas sp. A34and Xanthomonas oryzae pv. oryzicola BLS256etc. disappeared, Paenibacillus sp.3P02SAetc. generated; With the increase of the concentration of DO, Uncultured Enterobacter sp.clone LSSR150and Dechloromonas sp. A34etc. reduced or even disappeared, Paenibacillussp.3P02SA steadily growed finally. |
