The Antarctic krill living in the southern ocean is a great number of Marine crustaceans, which has7-8species basically. The Antarctic krill we usually said is Euphausia superba. The Euphausia superba containsabundant phospholipids resources, which is the biological enrichment library of phospholipids. The Euphausiasuperba phospholipids contain a lot of polyunsaturated fatty acids, which can prevent oxidation to delay ageing,protect liver, reduce the damage from body oxidation. At present the study and research of Euphausia superbaphospholipids resources is still relatively weak, therefore the studying of Euphausia superba phospholipidsprocessing technology and the physiological function experimental analysis in the laboratory condition is veryimportant to the development of our country, the Euphausia superba phospholipids resources industrydevelopment provides technical ideas and theoretical basis, and has important theoretical and economic value.Firstly,the phospholipid solvent extraction conditions from Euphausia superba organic is studied, bywhich we prepare the high purity Euphausia superba phosphatide, and then make the quantitative analysis of thecomponents and the corresponding fatty acid in phospholipid. In order to validate the anti-aging function of theAntarctic krill, we choose mice as the experimental object, observe the effects of ageing mice that fed withAntarctic krill phospholipids. The experiment provides the basis for the anti-ageing function of Antarctic krillphospholipids,which makes ideas for food and drug research and development.This paper mainly contains three parts:1.The extraction conditions optimization design of Antarctic krill phospholipidsThe Antarctic krill total lipids are extracted by organic solvent and purified by acetone, then the totalphosphatides are separeted. The experiment analysies the extracting-liquid composition, the proportion ofsolvent to material, the extracting time and other factors, by single factor experiment, and orthogonal test to getthe best preparation process of Antarctic krill phospholipids. The results shows that when the proportion ofchloroform to methanol is2to1, the liquid material is13times of krill tissue, and the extracting time is4h, theAntarctic krill extraction efficiency is the highest, which is about4.06%. 2.The phospholipids classes and fatty acid composition analysis of Antarctic krillWe choose the Antarctic krill cephalothorax for the materials, then according to the optimization of theextraction methods said in the above part we extract the Antarctic krill total lipid.The phospholipids classes areseparated and the elements are detected. Referring to GB/T5009-2003the water measurement method in foodhygiene inspection methods, the water in the Antarctic krill internal organs is78.95%. After separating the totallipid, we use acetone to isolate the phosphatide, and then concentrate on the one-way thin layer chromatographywith standard phospholipids as controls, which separates different phospholipids components. After which wemeasure the main components in the phospholipids by molybdenum blues colorimetry, and the result is as follow:phosphatidylcholine is59.40%, and20.12%is for phosphatidy lethanolamine; By gas chromatography we analyzethe fatty acid composition of total phosphatide at last.3.The anti-aging effect in mice of Antarctic krill phospholipidsIn order to validate the anti-aging effect of Antarctic krill phospholipids, we selects KM male mice in8weeks, and divide them into six groups: the Blank control group, the Negative control group, and the Positivecontrol group and the low, medium and high Antarctic krill phospholipids dose groups. The mice wereintraperitoneally injected with a dose of D-galactose for6weeks to make the ageing mice model.The micereceived phospholipids by intragastric administration simultaneously.After42days,the thymus index and liverindex were measured,the SOD,CAT,GSH-PX activities and MDA concentration were assayed,and the conditions ofskin were studied. The result shows that Antarctic krill phospholipids could reduce the level of MDAconcentration,increase the level of CAT,SOD,GSH-PX,delay the aging-process,and repair skin aging-damage. In aword, Antarctic krill phospholipids could delay aging.
