According the experiment of Streptomyces Shandongensis titer determination of exploration, we contrast the biomass method and turbidimetry and found that turbidimetry is superior. Final we just make turbidimetry as a measurement method of the determination titer of antibioticsTo get the overproducing strain,we just made for UV-licl mutagenesis, DES mutagenesis, UV-High permeability mutagenesis, UV-antibiotics mutagenesis and microwave mutation. Finally,we get the overproducing strain which is three times as much as the original production and the production is380mg/L.The optimization of the fermentaton medium we have got the optimal medium: Sweet potato starch20g/L, MgSO40.5g/L, FeSO40.01g/L, K2HPO41.28g/L soybean cake powder4.037g/L. And the production of antibiotics is1297.61mg/L3.4times as much as the original production. The experiment of Fermentation condition optimization, the production of antibiotics has got to1346.5mg/L. And the best condition:initial PH7.5, inoculation10%, Fermentation Temperature32"C We also get the precursor into fermentation medium, and it is a pity that we have not found a precursor that could increase the production. we also determine the residual sugar, dissolved oxygen and fermentation exhaust. We can monitoring antibiotics fermentation through these parameters.This paper design a Further optimization of the extraction conditions which based on former design. And the extraction effect have been improved, tasted for original rate3-4times, as the content of fermented liquid15%-20%.We also find the antibiotic of Streptomyces Shandongensis has anticancer properties, Which has has good antitumor activity to some tumour cells in vitro. But with the increase of the concentration of the antibiotic, antitumor activity has not substantial increased. And there also need a further study for the antitumor activity.
