The high F ratio oligo-peptide is one kind of small peptide mixture contain highbranch amino acid and low aromatic amino acid,which shows many physiologicalfunctions, such as adjuvant therapy hepatic encephalopathy, antifatigue,disintoxicating and so on. Because it has the unique amino acid composition and theimportant physiological function,receives high attention of food and the medicine dayby day. Corn gluten meal is a by-product of corn starch plant,and its protein contentcan reach60%-70%,Because of special smell and lack of Lysine and Tryptophan,itsphysics Characteristic and food functional o not very well.So far,it is hard to exploitand sold as feed cheaply.The programe aimed at the development of protein through preparation of highfischer ratio oligo-peptides from corn gluten meal,which has great significance ofprotein-depth development of basil to enhance the economic value of corn gluten mealand resource utilization. The main resultof study are as follows:1、Compared the effect of five kinds of pretreatment on the2709alkaline proteasehydrolysing the corn gluten meal. The best pretreatment process was determined assheae at the speed of12000r/m for5min,α-amylase treatment,and immediatelytreatment at121℃for30min.The hydrolysis rate of2709alkaline protease rised from11.4%to13.0%after pretreatment2、Through assaying9kinds of proteases hydrolysis the corn gluten meal, the2709alkaline protease and flavourzyme were selected as the best protease. The optimalconditions double proteases were analyzed by orthogonal test. the optimal parametersof the2709alkaline protease were concentration of enzyme4.0%pH9.0, temperature50℃, hydrolysis time2h; the flavourzyme were concentration of enzyme4.0%, pH7.0,temperature45℃, hydrolysis time3.5h. Proteases under the conditions of perillameal of up to26.3%degree of hydrolysis.3、Using activated carbon adsorb and separate aromatic amino acid,at the sametime to complete decolorization. Through assaying6kinds of activated carbon adsorbthe mixture of high F value few-peptide, and then selected TX-367activated carbonas adsorbent separation. TX-367activated carbon identified adsorb and separatearomatic amino acid parameters:the amount of activated charcoal10%(m/v), pH3.0,regular temperature,time2h. The Macromolecular polypeptide was removed by ultrafiltration and the best ressure for ultrafilting was0.28Mpa.The salt was remove byion exchange chromatography and the fit Flow velocity was8times of column volumepre hour, the rate of desalination should achieve85.37%.The yield of high F ratiooligo-peptide after after all the puration was42.62%.4、The anti-fatigue effects of high F ratio oligo-peptide on mice were studiedthrough different close. The results show that High F ratio peptide can ignificantlyenforces the swimming durability of mice, strongly degrade the content of blood lacticacid and urea nitrogen,increase the content of hepatic glycogen of post exercise mice.The High F ratio oligo-peptide has effective actions of raising exercise tolerance,promoting glycogen reservation、inhibiting the metabolism of lactic acid and ureanitrogen, and the anti-fatigue action is enhanced by increasing the dosage.
