Methanol is one of toxic compounds in the tequila. The research of reducing content ofmethanol in the tequila has great significance to consumers. At present, the report aboutsources and reduce of methanol is few in our country. So, this paper is mainly to study thesources of methanol in tequila with cane juice as main material and specifically carried on aseries of study work in composition of cane juice, strain and fermentation.1. Optimization of the preparation techniques of wort. The content of reducing sugar,protein and leaching rate could be regulated through the optimization of the preparationtechniques of wort, And the optimum preparation techniques were determined as follows:2hours steeping at55℃,1.5hours saccharification at65℃and half an hour thermal insulationat72℃. The headspace gas chromatography method with the optimized temperature and timein raising temperature procedure and velocity of carrier flow was established fordetermination methanol in the tequila.2. Adding glycine、compound pectinase and cellulase to the cane juice to study that theimpact of them to the content of methanol. The amount of compound pectinase and cellulaserequired for the pretreatment of cane juice were56U/L and60U/L;The cellulose in1L canejuice produced11.2mg/L the absolute methanol and83.9mg/L the relative methanol. Thestudy identified the linear response relationship between glycine and the yield of methanol,y=103.3x-4.5R2=0.983.3. Saccharomyces cerevisiae was mutated with three methods. The ability of producingalcohol and methanol was compared. Six mutant strains which the content of methanol lowerthan the original strain were screening by mutagenesis.They wereDES-0.2-3,DES-0.8-2,DES-0.8-2,ARTP-90s-1, ARTP-90s-3and ARTP-120s-3.The resultshowed that the yield of alcohol reached36.4%vol and increased by23.0%than the parentstrain and the absolute content of methanol was48.3mg/L and reduced by21.2%than theparent strain and the relative content was79.6mg/L and reduced by36.0%than the parentstrain after ARTP-120s-3strain fermentation.4. The optinal condition of producing methanol of ARTP-120s-3was decided by thecrossexperiment.the temperature was20℃, the time was6d and the initial amount was 4%.and the yield of methanol was172.9mg/Land reduced by16%than parent strain.5. The study showed that the yield of menthanol was low by200r/min28℃shakered4hbefore staic fermentation.the yield of methanol was lowest when the rapid fermentationachieved4d and the relative content of161.1mg/L and reduced by6.8%than parent strain.
