In this paper, micro-filtration membrane and ultra-filtration membrane in the separation of Qingzhuan tea water extracts and ethyl acetate extracts were studied. Through the analysis of the main active ingredients and the inhibitory activities of the separated parts on vitro metabolism enzymes, we expect to select the potent a-amylase and lipase inhibitors from Qingzhuan tea.1Study on the separation and purification of Qingzhuan tea water extracts using micro-filtration membrane.1.1Analysis of the main active components of Qingzhuan tea micro-filtration liqior.Useing0.45μm and0.22μm micro-filtration membranes filtering the Qingzhuan tea water extracts, Compared with Qingzhuan tea water extracts, the results showed that the contents of amino acids, soluble sugars, protein, catechins, flavones, tea pigment in0.45μm and0.22μm membrane filtrates had no significant change. The contents of polyphenols in0.22μm membrane micro-filtrates decreased significantly.1.2Effect on Qingzhuan tea extracting by micro-filtration liqior on vitro metabolism enzyme.Water extracts of the Qingzhuan tea,0.45μm and0.22μm membrane micro-filtrates had a-amylase inhibitory activity, each with IC5011.30mg/mL,11.54mg/mL and11.54mg/mL. After the treating of micro-filtration, the inhibition of α-amylase had no obvious enhancement.Micro-filtration liquor of Qingzhuan tea had inhibitory effect to lipase. IC50of0.45μm micro-filtrate was18.89mg/mL, its inhibitory activity was higher than Qingzhuan tea water extracts and0.22μm membrane filtrate, both IC50were greater than20mg/mL.2Study on the separation and purification of Qingzhuan tea water extracts using ultra-filtration membrane.2.1Analysis of the main active components of Qingzhuan tea ultra-filtration liqior.The ultra-filtration were under the conditions of0.05MPa,35℃for Qingzhuan tea0.45μm membrane filtrating liquior by using100KDa ultra-filtration membrane,50KDa ultra-filtration membrane,30KDa ultra-filtration membrane,10KDa ultra-filtration membrane,5KDa ultra-filtration membrane. In the single ultra-filtration experiments, concentration of100KDa ultra-filtration membrane could enrich the theaflavins and thearubigins in some extent. Concentration of50KDa ultra-filtration membrane concentrated soluble sugars, protein, flavones, thearubigins, chahe pigment. Concentration of30KDa ultra-filtration membrane had the higher contents of amino acids, the contents of polyphenols and chahe pigment decreased significantly. Filtration of10KDa ultra-filtration membrane enriched thearubigins, the contents of protein, flavones and chahe pigment decreased significantly. Concentration of lOKDa ultra-filtration membrane enriched soluble sugars and chahe pigment, the contents of thearubigins decreased significantly. The contents of thearubigins and flavones in Filtration of5KDa ultra-filtration membrane significantly declined. Concentration of5KDa ultra-filtration membrane enriched soluble sugars, polyphenols and chahe pigment. In comparison with different ultra-filtration membrane, the contents of catechins and thearubigins were higher in solution of100KDa ultra-filtration membrane. The contents of soluble sugars, protein, flavones, thearubigins and chahe pigment were the most abundant in concentration of50KDa ultra-filtration membrane. The contents of amino acids were higher in filtration of30KDa ultra-filtration membrane.The contents of polyphenols were the most abundant in concentration of5KDa ultra-filtration membrane.In the continuous ultra-filtration experiments, the contents of theaflavins were the most abundant in concentration of100KDa ultra-filtration membrane. Solution of50KDa ultra-filtration membrane could enrich the contents of amino acids and thearubigins, the contents of protein, flavonoids. catechins and chahe pigment were lower. Concentration of50KDa-100KDa ultra-filtration membrane enriched soluble sugars and chahe pigment, but the contents of thearubigins decreased significantly. Solution of30KDa ultra-filtration membrane enriched amino acids. Filtration of lOKDa ultra-filtration membrane could enrich thearubigins. Concentration of10-30KDa ultra-filtration membrane could concentrate the contents of amino acids; the contents of protein, polyphenols and chahe pigment decreaced significantly. The contents of flavonoids in concentration of5-10KDa ultra-filtration membrane decreaced. In comparison with different ultra-filtration membrane, the contents of soluble sugars, protein, flavonoids and chahe pigment were the most abundant in concentration of50KDa-100KDa ultra-filtration membrane. The contents of catechins and thearubigins were higher in solution of10KDa ultra-filtration membrane. The contents of polyphenols were higher in concentration of10KDa-30KDa ultra-filtration membrane. The contents of amino acids were the most abundant in solution of5KDa ultra-filtration membrane.2.2Effect on Qingzhuan tea extrating by ultra-filtration membrane on vitro metabolism enzyme.Qingzhuan tea single ultra-filtration liquor had the inhibitory activity of a-amylase and lipase. After the treating of0.45μm membrane micro-filtration, the components of different ultra-filtration liquor on a-amylase were weaker than before. The components with the most inhibitory activity on the lipase was the concentration of100KDa membrane ultra-filtration whose IC50was less than0.45μm micro-filtrating liquor’s, but the others IC50were greater than its.Qingzhuan tea continuous ultra-filtration liquor had inhibitory effect to a-amylase and lipase. The results showed that all the ultra-filtration liquor on a-amylase with IC50were higher than0.45μm micro-filtration membrane filtrating liquor. The components with the most inhibitory activity on the lipase was the concentration of100KDa membrane ultra-filtration with IC50of11.16mg/ml. The others with IC50were greater than0.45μm micro-filtrating liquor’s.3Study on the separation and purification of ethyl acetate fraction from Qingzhuan tea using ultra-filtration membrane.3.1Analysis of the main active components of ethyl acetate fraction ultra-filtration liquor.The ethyl acetate fraction was filtrated with5KDa ultra-filtration membrane under the conditions of0.05MPa,35℃. The results indicated solution of5KDa ultra-filtration membrane only enriched the contents of chahe pigment, in comparison with ethyl acetate fraction, the rest decreased significantly. The contents of polyphenols, flavones and chahe pigment of concentration of5KDa ultra-filtration membrane inhanced significantly, theaflavin content reduced significantly.3.2Effects of Qingzhuan tea ethyl acetate extracting fraction treating with ultra-filtration on vitro metabolism enzymeUsing5KDa membrane ultra-filtrating the ethyl acetate fraction, the concentration of ultra-filtration had the a-amylase inhibitory activity with IC503.88mg/ml and the filtrate had the lipase inhibitory activity with IC501.71mg/mL,each less than the ethyl acetate fraction’s.
