In this study, we investigated characterization,dye colorization ability and application of Clostridium Prazmowski spore laccase. Laccase produced by Clostridium Prazmowski is located on the bacillus outer wall protein. Clostridium Prazmowski was removed from the solid-sporulation media, washed by sterile deionized water, NaCl and SDS,then finally suspended in distilled water.Biological characteristics of the strains was studied, the optimum growth temperature and pH was28℃and7.0respectively.The strain can grow normally in the1%~5%salt concentration conditions. Clostridium Prazmowski reach the maximum growth rate at18h and can torlerate2mM copper ions.Spore laccase produced by Clostridium Prazmowski oxidized syringaldazine as its only substrate at optimum of pH7.0and70℃.Spore lacase activity was determined at20and100℃,and laccase activity could be determined at100℃. Spore laccase showed higher activity within a temperature range from40to70℃. The stability of spore laccase activity in organic solutions was Tween80>petroleum ether>methanol>Triton X-100>ethanol.Metal ions had great effect on spore laccase activity,most metal ions activated laccase activity,while Ag+、 Mn2+and Hg2+showed a strong inhibition effect.In the study of dye decolorization,orange I, alizarin red,crystal violet and RBBR could be decolorized by the spore laccase without mediator,while the decolorization of alizarin red,crystal violet and RBBR proceeded slowly.When mediators was added,the enzyme could rapidly decolorize alizarin red,crystal violet. and RBBR The results demonstrated the potential application of Clostridium Prazmowski spore laccase in the treatment of dye decolorization.Besides, this article also applies orthogonal experimental method to study the optimal decolorization conditions of spore laccase decolourizing four dyes, and shows the importance of the spore laccase in dye decolorization.Self-designed device was used in this paper,and spore laccase suspension,2,4-DCP solution, alpha-naphthol solution and mixed dyes was filled in the device resplevtively.Hence,we determine spore laccase reuse times. Experimental results show that the spore laccase of Clostridium Prazmowski has a good recycling.Spore laccase has great application potential the Bacillus laccase in the phenolic wastewater, as well as dyeing and finishing industrial wastewater treatment.
