This study focused on solvent-free synthesis of glycerol solution of1,3-diacylglycerol. Frist, finding out the highest activity of lipase through study of three different optimal reaction conditions. Second, according to the different nature of the lipase, emulsifier and ultrasound used in the synthesis reaction, to enhance the enzyme activity and increase of1,3-diacylglucerol production rate. At last, high performance liquid chromatograph was used to measure the synthetic samples.Three kinds of lipase Lipase8000, Lipozyme RMIM and Novozyme435the impact of reaction conditions factors are temperature, reaction time, substrate molar ratio and the content of lipase. TLC, column chromatography and HPLC/RID were used to detection of reaction products. The optimum conditions of Lipase8000:38℃, blend oil/glycerol(m/m):2:1, the content of lipase:3%,31h,1,3-diacylglycerol highest level of12.35%. The optimum conditions of Lipozyme RMIM:38℃, blend oil/glycerol(m/m):2:1, the content of lipase:4%,31h,1,3-diacylglycerol highest level of34.47%. The optimum conditions of Novozyme435:50℃, blend oil/glycerol(m/m):1:1, the content of lipase:8%,43h,1,3-diacylglycerol highest level of41.97%. The factors affecting emulsifier reaction are the kinds of emulsifier, dosage. The optimum conditions:polysorbate of15mg/mL,1,3-diacylglycerol highest level of45.53%. The ultrasonic treatment of Lipozyme RMIM, reaction factors:continuous ultrasound/stop ultrasound, total time, ultrasound power and method. The optimum conditions:60s/30s,30min,80w, ultrasonic between the reaction,1,3-diacylglycerol highest level of44.52%.The optimum conditions of synthesis of1,3-diacylgly:Novozyme435,50℃, blend oil/glycerol(m/m):1:1, the content of lipase:8%, polysorbate of15mg/mL.
