| This study used Aureobasidium pullulans which were mutagenized D1-11as rawmaterial, prepared pullulan zymotic fluid, preferenced the method of deproteinzation on zymoticfluid, and researched the influence of solubility, intrinsic viscosity, and researched the influenceof viscosity on pullulan mixed with gelatin, xanthan gum, sodium polymannuronate, carrageenan,guar gum etc.. Aight kinds of different de-protein methods were compared in this study, and thetechnology parameters of papain hydrolytic method were optimized; DEAE-52cellulose wasselected by the adsorption and resolution which was suited of purification on pullulan zymoticfluid, and optimized its technological parameter, the solubility, intrinsic viscosity of pullulan andthe influence of viscosity on xanthan gum etc.colloidal solution were studied, The main resultsobtained are as follows:1. Comprehensive comparison showed: papain was the suitest method ofdeporteinization on pullulan zymotic fluid.The result of the optimum parameters of papainhydrolytic method, which were optimized by response surface method, were: pH5.19, enzymesolution time2.21h, enzyme solution temperature49.08℃, amount of enzyme1.59%. Under thatcondition, the biggest protein removal rate was64.12%, the loss rate of polysaccharide was13.06%.2. Comprehensive comparison showed: DEAE-52cellulose was the suitest absorptionpurification on pullulan zymotic fluid.The results of the technique optimization of the dynamicdesorption on pullulan of DEAE-52cellulose were were: adsorption pH for8, adsorptionvelocity for medium speed, elution pH for7.0, the concentration of NaCl2.5mol/L, elutionspeed for medium speed. Under that condition, the desorption rate was86.13%.3. The results of pullulan solubility were: temperature had large effect on the solubility,relatively insoluble at lower temperatures, pH and salt concentration had less effect.4. Different effects will be produced different effects by crude polysaccharideconsistency, temperature, salt concentration, pH, heat treatment time, shearing, metalions on theviscosity of pullulan solution, its resistance to degradation performance was better.5. Either between the pullulan and several other polysaccharides to produce thephenomenon of thinning, but also with each other to produce thickening phenomenon. |
