Study On In Situ Expression Of Wnt Family Wnt4-like And FLNa In Skin Of Phrynocehalus Przewalskii

Abstract/Summary:
There is the capacity of renewable in the skin of Phrynocehalus przewalskii, but the molecular mechanisms of proliferative capacity are unclear. We studied the microstructures of the skin and scale sensory organ, observing the shape and distribution of the skin pigment, scale sensory organ and the mRNA levels of expression of Wnt family gene Wnt4-like and FLNa in the skin of Phrynocephalus przewalskii by the micrographic technique, scanning electron micrograph, HE staining, immunohistochemistry and in situ hybridization. The experimental results showed:(1) The shape of the scales and the structure of scale sensory organ have closely relation to the living habit and perception of the environmental stimulating factor;(2) There is a dermal corpuscle in the dermis of sense receptor which is similar to the feathers or hair’s Meissner’s corpuscle-like structure and inferred that the scale sensory organ may have the function of feeling mechanical stimulation and thermal radiation;(3) We have observed that Wnt4-like mainly expresses in the germinal layer cells of skin epidermis and scale sensory organ, but no expression in other cell layer (Oberhautchen, β-keratin layer, mesos layer, a-keratin layer);(4) By gene clone,we found that the sequence of Wnt4-like is highly similar to Wnt3a;(5) FLNa mainly expresses in the germinal layer cells of skin epidermis and some scattered cells in dermal tissue. These results can supply us reliable evidence to further research to Wnt signaling pathway and FLNa, which are involved in the development and regeneration of the Phrynocehalus przewalskii’s skin.
