As a relative protein member of action, palladin has been attracting more andmore attention, nowadays. Especially, it’s widely use in eukaryotic expression and theimportant role it played in cell, function as the construction of the cytoskeleton andsignal transduction in cell signaling pathways, for example. In this study, we justfocus on the influence of palladin on cell differentiation and relationship with theinvasiveness of tumor cells.In the thesis the influence of palladin on cell differentiation, we focus on thedifferentiation process of PC12(rat pheochromocytoma cells) to neural cell inducedby NGF (nerve growth factor). Experiment was designed from both positive andnegative aspects to prove the influence of palladin on the differentiation of PC12. Onthe one hand, we have constructed the palladin gene expression silencing vectorpGenesil-1-R-palladin-shRNA for silence PC12palladin expression, at the sametime, add NGF to the system, to investigate the inhibition of the palladin genesilencing on the differentiation process of PC12towards neural cell induced by NGF.On the other hand, we introduce palladin eukaryotic expressed pEGFP C3-MPForiginated from our lab into PC12, to detect the influence of overexpression ofexogenous palladin on the differentiation degree of PC12cells. Experiment result tellus that the overexpression of palladin could facilitate the conversion of PC12cells toneural cells, and silent expression of palladin could inhibit the cell differentiationprocess of PC12induced by NGF effectively, which proved the important rolepalladin play in the process of neuronal differentiation process necessarily andsufficiently.In the thesis the relationship of palladin and tumor cell invasion. We examinedthe cell invasion capacity and the basal expression level of palladin of MCF7, HeLaand HT1080, and found that the palladin expression levels were MCF7<HeLa<HT1080, the same trend could also be found in cell invasion capacity test. Knockdown of palladin could decrease the cell invasion capacity of HeLa and HT1080, butoverexpression of palladin exhibit no conspicuous effect on cell invasion capacity.This study focus on the effect of palladin on the differentiation of nerve cell and facilitation of cancer cell invasion capability, which enriched the principle system ofpalladin played in vivo. The chapter influence of palladin on cell differentiationprovide us new perspective on the nerve trauma reparation and the treatment ofneurodegenerative diseases. And in the chapter the relationship of palladin and tumorcell invasion emphasize the relation between palladin and the occurrence anddevelopment of tumor cells, which provide us more and more support on the study oftumor cell metastasis theoretically, and provide us a new target for the treatment ofcancer.
