In this paper, anthraquinone derivants (Alizarin, Alizarin-DA, Alizarin-DA-Fe)and acridines (Acridines orange, Acridines red, Neutral red) are selected assonosensitizers. Bovine serum albumin (BSA) is selected as a model protein for itstypical biological structure and important physiological function. The interaction ofBSA with those sonosensitizers and the sonodynamic damage of BSA underultrasonic irradiation were studied. If the biological macromolecules in the tumorcells are damaged by some sonosensitizers under ultrasonic irradiation, the whole cellwill die. In this study, the generations of Reactive oxygen species (ROS) weredetected by the method of Oxidation-Extraction Spectrometry. In daddtionm, in orderto searching for new sonosensitizers, the interaction of Amantadine Schiff-Bases(Amantadine-Salicylaldehyde, Amantadine-5-Chloro-Salicylaldehyde,Amantadine-o-Vanillin) with BSA is also study. This research is on the basis ofultrasonic irradiatetion activates porphyrins and porphyrin derivatives to damagebiological molecules. This paper not only proposes a kind of natural and nontoxicsonosensitizers for SDT, but also provides theoretical references for sonodynamictheapy to further explore the more effective sonosensitive drugs. This paper mainlyincludes the work listed as follows.1. The interaction between BSA and anthraquinone derivant (Alizarin,Alizarin-DA and Alizarin-DA-Fe) were investigated. The results showed that theseanthraquinone derivant could efficiently bind to BSA in aqueous solution.Synchronous fluorescence spectroscopy reveals that these anthraquinone derivantadopt different way to bind with BSA molecules. In addition, the damage of BSAhappened under ultrasonic irradiation in the presence of those anthraquinone derivantwere estimated by means of UV-vis and fluorescence spectra. Meanwhile, someinfluencing factors such as ultrasonic irradiation time, anthraquinone derivantsconcentration and ionic strength on the damage of BSA molecules were considered.At last, the generations of ROS under ultrasonic irradiation in the presence of those anthraquinone derivant were researched by Oxidation-Extraction Photometry.2. The interactions between BSA and Acridines (Acridines orange, Acridines red,Neutral red) were investigated by fluorescence spectra. The results showed that theseAcridines could efficiently bind to BSA in aqueous solution. Synchronousfluorescence spectroscopy reveals that these Acridines adopt different way to bindwith BSA molecules. In addition, the damage of BSA happened under ultrasonicirradiation in the presence of these acridines were estimated by means of UV-vis andfluorescence spectra. Meanwhile, some influencing factors such as ultrasonicirradiation time, acridines concentration and ionic strength on the damage of BSAmolecules were considered. At last, the generation of ROS under ultrasonicirradiation in the presence of these Acridines was researched.3. As a novel sonosensitizer, Amantadine Schiff-Bases were synthesized andcharacterized by infrared spectrum and elementary analysis. Fluorescence quenchingwas used to study the interaction of these Amantadine Schiff-Bases(Amantadine-Salicylaldehyde, Amantadine-5-Chloro-Salicylaldehyde,Amantadine-o-Vanillin) with BSA. The results show that these AmantadineSchiff-Bases can obviously bind to BSA molecules. Synchronous fluorescencespectroscopy reveals that these Amantadine Schiff-Bases adopt different way to bindwith BSA molecules. That is, the Amantadine-Salicylaldehyde, andAmantadine-5-Chloro-Salicylaldehyde are accessibility to tryptophan (Trp) residuesmore than the tyrosine (Tyr) residues, while the Amantadine-o-Vanillin is equallyclose to the Tyr and Trp residues.
