Cytokinins play many vital roles in plant development and physiology process. To be better understand the role of two component signaling system in maize, we analyzed TCS genes by using bioinformatics and molecular biology method in this study. The expression patterns of TCS components in the transgenic plants with both overexpressed and RNAi for ZmRR5gene were investigated. The results were showed as following:1. Using bioinformatics software, a total of48genes had been found to encode putative TCS genes, including11ZmHKs,9ZmHPs, and28ZmRRs (21A-type ZmRRs and7B-type ZmRRs).2. Based on phylogenetic analysis, the closed relationship of TCS genes were found between rice and maize. The duplication of these two-component system genes on the maize chromosomes corresponded to the clusters of these genes in the phylogenetic trees. The RT-PCR results showed tissue-specific expression in ZmRRs.3. After ZmRR5genes was obtained, the overexpression vector and RNAi vector of ZmRR5genes were constructed and transformed into maize mediated by Agrobacterium. Finally T2generation transgenic maize for ZmRR5overexpression and RNAi with single copy external gene were picked out by PCR and outhern blot examination system.4. Compared with B73, when treated with given concentrations of6-BA, the ZmRR5overexpression transgenic plants showed lower sensitive while the ZmRR5RNAi transgenic plants showed more sensitive. When treated with given concentrations ofABA and2,4-D, there was no difference betweeen B73and transgenic plants. This phenomenon illustrated that ZmRR5is only Cytokinin response regulator, not other hormones response regulator.
