The WRKY proteins are a superfamily of transcriptional factors which is widelydistributed in plants. The family members regulate expression of related genes by bindingspecifically to the DNA sequence called W box, so as to regulate the process ofsenescence and the response of plants to many kinds of stresses.TaWRKY53c is a WRKY gene cloned previously in our laboratory. The presentresearch aims to define the role of TaWRKY53c in different signaling pathways byanalyzing expression changes of TaWRKY53c in wheat seedlings treated with differentstresses or phytohormones using real-time quantitative PCR and to obtain TaWRKY53cover-expression of transgenic tobacco plants in order to study its function. Wheatseedlings are treated with eight kinds of stresses and phytohormones(PEG、NaCl、ABA、SA、H2O2、ETH、MeJa、cold) for different period of time(1h、3h、6h、12h) for the analysisof relative expression of TaWRKY53c.For the plants treated by the eight kinds of stressesand phytohormones, the relative expression of TaWRKY53c at different times differed in acertain degree compared to that of control. Therefore, it is suggested that TaWRKY53cmaybe involved in the response of wheat to the eight kinds of stresses and phytohormonesto some extent.TaWRKY53c over-expression vector SN1301-TaWRKY53cOX wasconstructed by inserting ORF sequence of TaWRKY53c cDNA into the plasmid SN1301under the control of CaMV35S promoter. SN1301-TaWRKY53cOX was transformed intotobacco by Agrobacterium-mediated transformation method. Seven regenerated plantswere obtained, and three positive transgenic plants were confirmed. In conclusion, thepossible signaling pathway TaWRKY53c involved was preliminarily defined. TheTaWRKY53c over-expression transgenic tobacco plants provided useful materials forfurther function analysis of TaWRKY53c.
