Exploration and development of marine drugs has become one of the keydirections of the marine biological research。 Looking for the active substancefrom the rich marine resources which can be used for new medicinal value andhealth functions, has yet become the new hot spot。 The study found thatchitosan and its derivatives can perform a variety of biological activity, suchas biocompatibility,antimicrobial activity to enhance immunity,and inhibitionof tumor cell growth and physiological function, etc。 This project,on thisbasis, intends to study the proven regulatory role of chitosan derivatives onImmune injured mice, to determine whether it can be used as an immuneregulator for enhancing the immune system。Chitosan is a natural the polycationic biological polysaccharide, widely usedin biology。 Studies have shown that chitosan holds a lot of biological activitiesand medicinal value, such as good biocompatibility, non-toxic, inhibitingGram-negative bacteria and positive bacteria。 Also it can effectively reduceblood pressure, blood glucose and blood lipids and cholesterol in the liverand serum。 In addition, chitosan could promote the antibody formation andstrengthen immunity and disease resistance。But the defect of water-insolublenature limit its application as a health supplement or drugs。 Itsbiocompatibility and water solubility can not meet the injection or oral conditions, restricting In-depth study and limiting practical application of itsbiological functions。 The application of modified chitosan as a materialimpacting on immunocompromised mice, can better the development andexploration of biological activity, medicinal and health care functions ofchitosan。To further explore the role of chitosan on immune function,in this project,weuse carboxymethyl chitosan,50%degree of deacetylation of chitosanoligosaccharide, chitosan oligosaccharide as experimental materials。Immunocompromised mice model is attained by injecting Cy intointraperitoneal。 The experiment use conversion of mouse body weight,spleen index, thymus index, as well as lymphocyte proliferation, NK cellkilling activity and macrophage phagocytic activity changes as an indicator toinvestigate the immunomodulatory effects of chitosan derivatives onimmunocompromised mice, in order to provide the experimental basis for theapplication of chitosan derivatives as health care or drug to enhance immunity。In this paper,applying carboxymethyl chitosan,50%degree of deacetylationof chitooligosaccharides and chitooligosaccharides as experimental material,implementing the intragastric administration of different doses on immuneinjury mice to measure the body weight, spleen index, thymus indexmeasurements, as well as lymphocyte proliferation, cytotoxic activity of NKcells, macrophages activity。The experiment observed the immunomodulatory effects of chitosan derivatives on Cy induced immune-injured mice。The results indicated that thesingle Cy injection significantly inhibited the tested subjects’ spleen weight,spleen cell activity, NK cell activity and macrophage activity。 But theseindexes were restored to the normal control group of mice or near the level ofnormal group by gavaging carboxymethyl chitosan and50%degree ofdeacetylation of chitosan to immunocompromised model mice。 Theexperimental results showed that a certain concentration (300mg/kg) ofcarboxymethyl chitosan and50%deacetylated chitosan can dramaticallyimprove immune function caused by Cy。...
