Objective:We choose the Han nationality and Chinese Koreans in Yanbian as the study subjects. The goal of the research is to explore the relations-hip between the transcription factors7-analogues2(transcription factor7-like2, TCF7L2) gene single nucleotide polymorphisms (SNPS) rs7903146(C/T) and the metabolic disease such as the type2diabetes mellitus (T2DM).Method:We utilized Polymerase Chain Reaction-Single Strand Conformation Polymorphism (PCR-SSCP)and sequencing to test the gene of TCF7L2SNP rs7903146. We examined380Korea minorities and312Han people in the Yanbian region of north-eastern China. Among those209healthy people were used as control. The volunteers were further subdivided into four groups:the T2DM (89volunteers,42Korea minorities and47Han people respectively); the Metabolism Syndrome (MS) group (232volunteers,88Korea minorities and144Han people respectively); the Essential hypertension (EH) group (62volunteers,30Korea minorities and32Han people respectively) and the control group. All the data were gathered with the volunteers in fasting conditions. We tested the basic physical signs, such as:blood pressure, height, weight, waistline and hipline, and Plasma Adiponectin (PA), insulin (ISN), blood fat (TC,TG,HDL-C and LDL-C), Fasting-Plasma Glucose (FPG) and Body Fat Ratio,Application homa-ir method for calculation of insulin resistance index (IRI). Using case-control comparison analyzing, we analyzed the correlation between genotype frequencies, alle frequencies and diseases occurrence and clinical standars.Results:1. The frequency of the allele of the rs7903146TCF7L2(C/T) and the genotype are in line with Hardy-Weinberg balance (p>0.05) in Korean and Han. Because the sample is balanced, so it can be researched as represent the group samples;2. The gene frequency distribution of the TCF7L2gene rs7903146polymorphism in controls between the Korean and Han in Yanbian area:T allele frequency of Han Korean-Chinese (2.2%and0.4%) is higher, but this was not statistically significant (χ~2=2.90,p=0.09), and the frequencies of3types of genotypes in the Chinese Koreans are lower than that of the Han nationality (CC:99.2%and96.6%, CT:0.8%and2.2%, with1.1%TT:0), but it have no sense of statistics (the chi-square=2.09, p=0.35), but have no sense of statistics either(χ~2=2.09,p=0.35).3. The case-control correlation analysis results of the rs7903146Polymorphic:(1) Compare the T2DM group and the normal control group:All samples case-control more dominant gene type [i.e.(CT+TT) the sum] frequency are5.6%and0.19%respectively no significant difference (p>0.05), T allele frequency T2DM group (3.4%) than the control group (2.0%) have higher trend (the chi-square=3.26, p=0.07, OR=0.35).The genotype frequency and allele frequency have no sense of statistics significant difference between the Pure T2DM group and the normal control group (p>0.05), whether put the Han nationality and the Chinese Koreans together nor respectively;(2) Compare the MS group and the normal control group:All samples are analyzed with case-control method and it showed that the dominant allele type (CT+TT) frequencies in MS group are significantly higher than normal group (5.6%and1.9%,%=6.53, p=0.04, OR=0.42), but T allele frequency in pure MS group are the same as that in the normal control group (2.8%and1.2%,χ~2=2.84,p=0.09). The Han nationality and the Chinese Koreans are layered for the further study, and we found that in the Korean group the dominant genes type (CT+TT) frequencies of MS group have higher trend than that in the control group (4.5%and0.08%,χ~2=2.98,p=0.08, OR=0.49), but in the Han nationality with MS that have no significant difference from the normal control group (3.5%and2.2%, χ~2=0.57, p=0.45).(3) The Comparison between the controls and the Simple EH group:Compared with the controls, the dominant genes type (CT+TT) frequencies of the patients were significantly higher (9.7%and1.9%,χ~2=10.38, p=0.01), and all the same the T allele frequencies of EH group are also significantly higher than the normal control group (4.8%and1.2%, χ~2=6.38,p=0.01), and we found that positive results only in the Korean through further analysis, the frequencies of dominant genes (CT+TT) in EH patients are significantly higher than in the control group (10%and0.8%,χ~2=7.77,p=0.00). T allele frequencies of5.0%are also significantly higher than the normal control group0.4%,(5%and0.4%,χ~2=7.67,p=0.01). But the Han nationality dominant gene type (CT+TT) frequencies have no difference with normal control group (10%and3.4%,χ~2=3.65, p=0.16), and the T allele frequencies have no statistically significant (5%and2.2%,χ~2=1.19,p=0.28);4. The correlation-comparison results of rs7903146polymorphism of genotype and the clinical indexes:There were no difference between genotypes of normal population in Yanbian area and this study in clinical indicators of FPG, BMI, the TC, TG and HDL-C and LDL-C, the percentage of body fat, INS and PA (p<0.05);5. The comparison results of TCF7L2gene rs7903146polymorphism between races: The genotype frequency of CT and TT among Yanbian Korean are0.008and0, lower than the Yanbian Han0.022and0.011, the Han of Chinese Beijing (HCB)0.047and0and Japannese Tokyoites (JPT),0.047and0.012, there is no significant statistically difference (p>0.05), while there is statistically difference that the T allele frequency of the Yanbian Han is0.004, which is significantly lower than that of HCB (0.023) and JPT (0.035)(p=0.04). But there is no difference with the Yanbian Han (0.022)(p=0.09); the TCF7L2gene rs7903146polymorphism genotype frequency and the allele frequency of Asian populations are significantly lower than that of the European (CEU)(CC=0.549, C=0.345, TT=106, T=0.279) and the African (YRI)(CC=0.0.531, CT=0.425, TT=0.044, T=0.257)(p<0.01);6. The associated-analysis results of the clinical features of the Korean and Han general population in Yanbian Region:The results of plasma PA that Han was significantly higher than Korean (p=0.04), and there are trends that BMI of Han is higher than that of the Korean (p=0.07), other clinical indicators of FPG, BMI, and the TC, TG and HDL-C, LDL-C, percentage of body fat and INS had no statistical significance (p>0.05);7. Results of Case-control Clinical index association analysis:(1)A11samples case con-trol the correlation analysis:The results show①he level of TC and LDL-C in T2DM, MS and EH group are largely higher than in normal group (p<0.01); but the values of BMK TG、Body fat percentage and PA between those groups were no differences;②the values of BMI、TC、TG、LDL-C、and body fat percentage are significantly larger than for controls (p<0.01),PA are significantly higher than for controls (p<0.01). No differences of HDL-C and INS were observed in the MS and control group (p>0.05);﹖he level of TC and LDL-C in EH group were higher than controls (p<0.01), but no differences for the FPG、BMI、TG、HDL-C、body fat percentage、IRI and PA are observed in the two groups (p>0.05);(2)Case-control correlation analysis results of clinical indicators of normal population of Yanbian area Korean and Han:T2DM group FPG, LDL-C, TC are higher than those in the control group (p<0.01), and the rest of the clinical index no difference (p>0.05); And in MS group of FPG,BMI, TG LDL-C significantly higher than those in the control group (p<0.01), and the other clinical indicators are no difference (p>0.05);(3)Case-control correlation analysis results of clinical indicators of normal population of Yanbian area and Han:In the Han people of T2DM FPG, LDL-C, TC are higher than those in the control group (p<0.01), and the rest of the clinical index no difference (p>0.05); And MS group of FPG, BMI, TG, LDL-C significantly higher than those in the control group (p<0.01), IRI also higher than those in the control group (p<0.04), PA are significantly higher than for controls (p<0.01)Conclusion:1.The polymorphism factor T of the TCF7L2gene rs7903146does not have a relation with the T2DM of the Korean nationality and Han people in Yanbian. 2.The polymorphism factor T of the TCF7L2gene rs7903146is the risk factor of MS and EH, the risk of the Korean nationality with T(CT+TT) infected by EH is0.35times than those with CC.3.TCF7L2gene rs7903146polymorphism genotype frequency and T allele there are racial differences, there are also ethnic differences, China’s Yanbian Korean T allele frequency of rare genes.4.There is a nationality difference in the content of PA in the blood,in which the Chaoxi an nationality than Han people. The content of PA in the blood is in negative correlati on with the body constitution.5. Plasma PA the ethnic differences exist, plasma PA and BMI negatively, low plasma PA is the risk of deeloping MS factors.6. TC、LDL-C are the dangerous factors of the infection of T2DM、MS and EH.
