Reduced glutathione is small molecule thiol compounds, in the cells of living organism with many bioactivities including radical scavenging, heavy metal detoxification or accumulation. Selecting edible plants with abundant glutathione is applicable for the heath care and accumulation of heavy metal.This research changed the traditional extraction method and used new method which breaking the cell and deriving were accomplished simultaneously. It could avoid the thiols being oxidized. Using5,5’-dithio-bis-nitrobenzoic acid(DTNB) as derivatizing reagent could determine the total amount of thiols. Using the o-phthaladehyde(OPA) as derivatizing reagent could determine the amount of glutathione in the samples with Standard addition method and Standard curve method. Finally, the amount of cysteine can calculate using the subtraction.Using the MALD-TOF mass spectrometry determined the small molecule thiols. Results showed that the sample cells had glutathione and cysteine. This research played an important role in the method of analysing active materials in the cells.
