Preliminary Functional Analysis Of F-box Family Gene FOA1in Arabidopsis

Abstract/Summary:
Arabidopsis gene FOAl belongs to F-box family. Prediction were done using1500bp upstream ATG nucleotide sequence by software, found that there were many hormone response elements. However, reports about this gene were limited to bioinformatics results up to now. This research mainly analyzed the relationship of FOA1and ABA, GA. The main research results were as the following:Firstly, two FOA1over-expressed transgenic lines FOA1ox1and FOA1ox2. were obtained by the Gateway Cloning and agrobacterium mediated immersing inflorescence methods, simultaneously, FOA1T-DNA insert mutants were bought from ABRC, then T-DNA insert homozygous mutant foa1was successfully identified. Wild type Arabidopsis were treated respectively with exogenous ABA, NaCl, GA3, MeJA, IAA and blue-ray for different time periods, show that the FOA1gene expression were regulated by ABA, NaCl, GA3, MeJA, IAA and blue-ray. FOA1expressed in every organ, but expressed higher in flower and root, indicate that FOA1might mainly function in root and flower. Secondly, under ABA treat condition, the statistic data of seed germination, root elongation, stomata closure, proline accumulation of wild type, foal FOA1ox1, FOA1ox2shown that foa1was more sensitive to ABA, while FOA1ox1, FOA1ox2were less sensitive to ABA, a series of ABA signal transcription factors expressed higher in foa1mutant than in wild type, however the expression of ABA and stress response gene were lower foal than in wild type. These results illustrate that FOA1might function negatively in ABA signal transduction pathway, meanwhile, it may participates ABA signal transduction by adjusting the degradation of transcription factors related to ABA and stress response. Lastly, through the germination rate, root length, and hypocotyl elongation statistics of wild type, FOA1ox1, FOA1ox2, foa1treated with different GA3or PAC concentrations, demonstrated that foa1was more sensitive to GA3and PAC, while FOA1ox1, FOA1ox2was less sensitive to GA3and PAC. When treated with GA3plus PAC, FOAlox1, FOAlox2presented more sensitivity to GA3, foal remain the same sensitivity to GA3as wild type. By means of QRT-polymerase chain reaction (PCR) method, the expression level of some GA synthetic or metabolic pathway key genes in wild type and foal mutant were detected, the results showed that the expression of early synthetic gene and metabolic gene were lower in foal mutation, explained that mutant foal reduced GA synthesis, FOAl may positively control GA synthesis or signal transduction pathway.FOAl protein have F-box domain, thus may be functions as an SCF complex component to regulate GA synthesis, and participates in ABA signal transduction via affecting the degradation of transcription factors related to ABA and stress response, nevertheless these results still need further research.
