Common histological Hematoxylin&Eosin staining method, Real time quantitativePCR and immunofluorescence staining were used to study the morphology and expression ofpolarity protein Crb1in mouse ovary, uterus, testicle, epididymis at different postnataldevelopment stages.The results showed that: With ontogeny, growing follicles were more and more, and alarge quantity of antral follicles formed at4th week; at6th week, there were maturescentfollicles on the surface of ovary; corpus lutea could be seen at8th week; at12th week, themorphological character of follicular atresia was obvious. Vessels and interstitial glands in theovary medulla were became more and more with the mouse development. The lumina of uteriand uterine glands were gradually expanding; the uterine endometrial epitheliums werecontinuous thickening; and the uterine glands became more and more developed. At4th week,cilia towards lumen could be obviously seen, and they would be more and more developed.In mice ovaries, Crb1were mainly located in the membrane and cytoplasm of interstitialgland cells, luteal cells and thecal cells at different developmental stages.The level of Crb1expression in these cells were as below: interstitial gland cells> luteal cells> thecal cells. Theexpressions of Crb1protein and mRNA increased with the ovary development, and thendeclined. In mice uteri, Crb1was mainly located in the membrane and cytoplasm of uterineendometrial epithelial cell and uterine endometrial glandular epithelial cell. The expression ofCrb1protein and mRNA firstly increased and then declined with the development of uterus,and reached a maximum level at8th week.In mice testicle, Crb1were mainly distributed in the membrane and cytoplasm of bothseminiferous epithelium cells and leydig cells, and the positive reaction in leydig cells wereobviously stronger than that in spermatogenic cells. The expression of Crb1protein andmRNA in testicle firstly increased and then declined with the development, which reached aminimum level at2th week, and reached a maximum level at8th week. In mice epididymides,Crb1protein is mainly distributed in apical membrane of ductus epididymidis epithelial celland has weakly expressed in the tail of sperm. The expression of Crb1protein and mRNA inepididymis firstly increased and then declined with the development, which reached a maximum level at4th week and kept a low level at other stages.It suggested that Crb1was related with the establishment and maintenance of epithelialcells of reproductive organ, and had potential effect on the synthesis and secretion of steroidhormone.
