Urostyla sp. Cells of rupture of membranes based on the laboratory method, toimprove the experimental method, according to the characteristics of Parameciumbursaria vital signs, observed the internal structure of Paramecium bursaria under theelectron microscope.The structure under pellicle of Paramecium bursaria was observed by scanningelectron microscopy (SEM) and transmission electron microscopy (TEM). In thepreparation of sample for SEM, we used KMnO4to dissolute the P.bursaria cytoplasmic,so leave the independent cell membrane and the attachment structure of the innersurface. Then we turned the membrane to reveal the structure and the morpha of itspellicle inner surface. Results are as follows:1. There are full of like quadrilateral pellicular alveolus on the inner suface ofpellicle. They inward bulge, neatly arraged, and their size is about0.6×1.2μm.2.Several layers of retiform fiber covered the pellicular alvrolus and wrapped trichocystsand other intracellular structures. These fibers include kinetodesma, postciliarymicrotubules, transverse microtubules and various cytosketeton fibers.3. There are alarge number of spindle-like structures and a small amout of rod-like structures in cell.Compareing the results with TEM, they are two kinds of trichocysts. One is spindle-like.It’s about3μm long. And the end has a little tip that was showed by TEM. The other isrod-like about8-10μm. Under the TEM, there are stripes on its’s surface. The two kindsof trichocysts all distribute between two pellicular alveolus. In addition, many symbioticchlorella in the P. bursaria were observed by SEM.Their diameter is about3.5μm.4.The cytopharynx inwarding bulge was also observed in the P. bursaria by SEM. It wascoverd by multi-fibers which woven into a basket-like structure. Around the basket,there are many spherical particles. The diameter of the uplift cytopharynx is about7μm.It is the first time that observed the structure of Paramecium bursaria by both ofscanning electron microscopy (SEM) and transmission electron microscopy (TEM), tofill the research gap, added new informations for the taxonomic study.
