Population dynamic is the object that ecological scientists attempt to understand. This sbuject includes many aspects such as spread of individuals in a certain animal population, variation of population size, sexual structure of population and so on. Currently, the studies to population dynamic mainly focuse on some small wild lives which contains insect and rat species. In this field, however, the studies about bigger ones are rarely reported. The first step we should take is to identify animal indivaduals accurately, which is the basic requirement for the study of population dynamic. So individual identification is of great importance for the future research which is necessary to understand those who needs protection immediately. Individual identification technology has now been used wildly in many respects. It contains VNTR (Variable number of tandem repeat), RAPD (Random amplified polymorphic DNA), SSR (Simple sequence repeat, Microsattlite), STR (Short tandem repeat) and so on, in which SSR has been regarded higher arability to be a individual identification system for the deep research.Individual identification of wild animals is one of the most important subjects in field of population dynamic research. Currently, microsatllite DNA fingerprinting technology has been used frequently and successfully on many kinds of animals, and that makes the application of this type of fingerprinting technologies available to our study. We constructed identification system using8microsatllite loci, by which31black muntjacs were identified from141fecal samples correctly. The analysis of software CERVUS3.0and POPGENE1.21showed that the identification system had strong discrimination power:Discrimination power0.938per loci (DP),0.999in total (CDP), the mean observed number of alleles(Na) was8.875, the mean effective number of alleles (Ne) was6.375, and mean heterogosity (He) was0.829. In this paper we, meanwhile, primarily analyzed the sexuality ratio of those individuals that had been identified. For the importance that the changes of sexuality ratio in population could exert significant impact on population growth, density and dynamic to some extent, sexual identification is also a key object in our study. SRY gene was used to analyze those individuals sexuality. The results showed that there were19males and12females based on agarose gel electrophoresis data. From what difference between the both sexuality could be easily found. In addition, we testified the result of sexual identification by sequencing. It showed that mean similarity with the subject suquence was above95%.Our study, the further step after exploring the method of fecal DNA extraction, was to keep well track of the enigmatic organism, balck muntjac, and provide some direct evidence on studying of this kind. It should be reference to others alike.
