Non-coding RNA is a single-stranded ribonucleic acid, it is transcript by DNA but doesn’t encode protein. Already known as transporter RNA, ribosomal RNA, and some found late non-coding RNA, such as microRNA, small nucleolar RNA and small nuclear RNA. The study found, non-coding RNA in organisms plays an important role:regulation of gene expression, participation in the RNA shear and modification. In the present stage, high-throughput sequencing technology is one of the most important means.People always focus on one specific non-coding RNA, this experiment focus on all the non-coding RNAs between100-200nt. The templates are cortexes of human, chimp, rhesus and mice. Experimental innovation is that to establish a100-200nt cDNA library take illumina’s standard protocol as example, and sequence the library for high throughput sequencing.Finally, we map the reads to the reference genome, get the annotated non-coding RNA, compare the sliding window result to annotated information, finally, we set a strict cutoff of the sliding window, We discover59new non-coding RNA, there are20kinds of which have a large number of expression (sequence>50), the found of new non-coding RNAs are supplement to non-coding RNA library, lay the foundation for the future experiment. And4non-coding RNAs are high conservative. And we make some family comparison of homologous genes such as SNORA29, it express in other species not human, this particular gene may be focused later.
