microRNAs are a group of key non-coding RNAs recently discovered in cells. They play an important role as a post-transcriptional regulation factors in gene expression of higher eukaryotes. microRNAs exist widely in creatures and function as regulators, thus research into their regulation becomes more and more significant.mir-7and mir-153link respectively with individual development and some cancers. mir-7protects individual’s healthy development in alternating conditions, which earns its crucial position in stability of molecular networks, mir-153down-regulates expression of gene APP and APLP2, which are the main malgenic genes in Alzheimer’s Disease, and decrease the formation of degradation fragment. This paper focuses on mir-7, mir-153and their interactions with target genes.In our experiment, mir-7and mir-153gene were constructed into the pmR-mCherry, their13target genes into pGL3-Control. Next, constructed plasmids were transfected into293T cells to measure the expression amount of target genes. During transfection, we employ one of the targets ARID4A for the ratio of different miRNA and mRNA.Theoretically, mathematical model were used for the interaction of mRNA and miRNA, mRNA stability as well. This paper respectively calculates the α, ratio of between microRNA and mRNA, and γ, binding ratio of microRNA and mRNA. Experimental data tallies with theoretical data after comparison, which in turn offers a theoretical model in describing interactions between miRNA and mRNA.All data were collected in Professor Ge Shan’s lab, College of Life Sciences, USTC. The ownership of experimental data belongs to Professor Ge Shan’s group.
